Parylene peel-off arrays to probe the role of cell–cell interactions in tumour angiogenesis

Abstract: Microenvironmental conditions impact tumour angiogenesis, but the role of cell-cell interactions in modulating the angiogenic capability of tumour cells is not well understood. We have microfabricated a peel-off cell-culture array (PeelArray) chip to spatiotemporally control interactions between tumour cells in a large array format and to analyse angiogenic factor secretion in response to these conditions. The PeelArray chip consists of a polyethylene glycol (PEG) treated glass coverslip coated with a parylene… Show more

“…For example, O 2 -dependent changes in cell proliferation may alter cell-cell interactions via direct cell-cell contact and altered paracrine signaling. 38,39 By covalent coupling of cell-cell adhesion molecules (e.g., cadherins) to alginate or utilization of alginate-based matrices to recreate biologically inspired delivery profiles of soluble factors (e.g., gradients), it may become possible to study the role of these cues under defined O 2 and 3D culture conditions. Finally, while ambient O 2 is commonly employed in cell culture work, physiological tissue O 2 levels are significantly lower.…”

“…Several approaches have been designed for this purpose, e.g., inkjet cell printing (3)(4)(5)(6), surface engineering (7)(8)(9)(10)(11)(12)(13)(14)(15), and physical constraints (16)(17)(18)(19)(20)(21)(22)(23). However, finding a method that completely satisfies the above requirements remains a challenge.…”

“…26 However, during tumorigenesis these transmembrane proteins are often downregulated, 27,28 which can induce a change in cell-cell interactions as well as impair tissue polarity, both of which can regulate the angiogenic capability of tumor cells. 29,30 Altered microenvironmental concentrations of pro-angiogenic factors, in turn, control the formation of intercellular junctions between endothelial cells by modulating expression and signaling of VE-cadherin. 31,32 These changes are directly relevant to tumor vascularization since dysfunctional VE-cadherin signaling ultimately promotes invasion angiogenesis and blood vessel permeability.…”
