PAH biomarkers for human health risk assessment: a review of the state-of-the-art

Abstract: Polycyclic aromatic hydrocarbons (PAH) are widely distributed in the environment, and some are carcinogenic to human beings. The study of biomarkers has helped clarify the nature and magnitude of the human health risks posed by such substances. This article provides a review of the state-of-the-art on PAH biomarkers for human health risk assessment and also discusses their applicability within the context of environmental management in Brazil. The article discusses the methodologies for determination of some b… Show more

“…There are no other studies reporting CYP1A1 * 3 frequency in AML patients [3][4][5][6][7][14][15][16][17][18]. Thus, by the multivariate analysis our data showed that CYP1A1 * 3 increases the risk of AML 4-fold.…”

“…PAH oxidation generates epoxide which is a very reactive compound that binds covalently DNA and unless repaired makes DNA adducts. The formation of DNA-adducts by electrophilic metabolites is generally regarded as one of the earliest steps in PAH carcinogenesis [3]. At present data, 11 CYP1A1 alleles have been reported; however, several are very rare and of unknown functional significance (a complete description can be found at http:// www.cypalleles.ki.se/cyp1a1.htm).…”

“…The vast environmental distribution of PAHs results in a great deal of human exposure to them. Also because of their lipophilic nature, PAHs are easily absorbed and distributed in the whole human body (Franco et al, 2008). PAHs are metabolized by xenobiotic metabolizing enzymes, which convert xenobiotic compounds into more hydrophilic and polar metabolites for their easier excretion in human body fluids.…”
