Oxidative Stress-Induced DNA Damage and Apoptosis in Clove Buds-Treated MCF-7 Cells

Abstract: In recent decades, several spices have been studied for their potential in the prevention and treatment of cancer. It is documented that spices have antioxidant, anti-inflammatory, immunomodulatory, and anticancer effects. The main mechanisms of spices action included apoptosis induction, proliferation, migration and invasion of tumour inhibition, and sensitization of tumours to radiotherapy and chemotherapy. In this study, the ability of clove buds extract (CBE) to induce oxidative stress, DNA damage, and str… Show more

“…[ 54 ] We further determined cell demise and apoptotic cell decease happened in TE‐13 cells in a time‐dependent manner by clove oil, which holds eugenol with 80% cell death while DU‐145 cells displayed negligible cell death. [ 55 ] The same results were observed in propidium iodide staining also in which Eu‐Pi/PHB‐PEG‐NC (5–7.5 μM) demonstrates red fluorescence of apoptotic cells, which is displayed in Figure 8. The control displays a normal appearance without any red fluorescence.…”

“…Pro-apoptotic activity of ERK has been reported previously. Aberrant ERK activation by neurotoxic Aβ 42 peptide leads to apoptosis in neurons [ 63 ], and prolonged activation of ERK beyond 24 and 48 h was associated with apoptosis induced by clove bud extracts in MCF-7 cells [ 64 ]. There are also reports showing that ERK activation by oxidative stress is associated with apoptosis in renal [ 65 ] and fibroblast cell lines [ 66 ].…”

“…Kumatakenin, a flavonoid isolated from cloves, induced apoptosis by enhancing the activity of caspase‐3, caspase‐8, and caspase‐9 in human ovarian cancer cells (SKOV3 and A2780) and in tumor‐associated macrophages (TAMs; Woo et al., 2017). Clove bud extract‐induced apoptosis, DNA damage, and oxidative stress in MCF 7 (breast cancer) cells (Kello et al., 2020).…”
