Osteogenic cell response to calcium aluminate‐based cement

Abstract: CAC+ supported the acquisition of the osteogenic cell phenotype in vitro, rendering this novel material a potential alternative to MTA in endodontic procedures. Further in vivo studies are needed to verify if the beneficial in vitro effects of CAC+ on osteoblastic cells correspond to an increase and/or acceleration of bone repair in the periapical region.

“…After washing in PB, cells were permeabilized with 0.5% Triton X‐100 in PB for 10 min and submitted to epifluorescence protocol as described previously (Castro‐Raucci et al . ) using Alexa Fluor 488‐conjugated phalloidin (green fluorescence, 1:200, Molecular Probes, Invitrogen, Eugene, OR, USA) for actin cytoskeleton labelling and 300 nM 4′,6‐diamidino‐2‐phenylindole, dihydrochloride (blue fluorescence, DAPI, Molecular Probes) for cell nuclei staining. The samples were examined under epifluorescence using an AxioImager M2 Zeiss light microscope (Carl Zeiss Inc., Oberkochen, Germany) outfitted with an AxioCam MRM digital camera (Carl Zeiss Inc.).…”

“…Hidroxyapatite-ZnO composites containing 10 wt% or lower ZnO, exhibited the ability to support cell adhesion and their proliferation 40 . Increased cell viability, proliferation and ALP activity were also observed for CAC exposed osteogenic cell culture, which was partially attributed to the Ca 2+ levels released by this cement 41 .…”
