Organic Salts of Pharmaceutical Impurity p-Aminophenol

Abstract: The presence of impurities can drastically affect the efficacy and safety of pharmaceutical entities. p-Aminophenol (PAP) is one of the main impurities of paracetamol (PA) that can potentially show toxic effects such as maternal toxicity and nephrotoxicity. The removal of PAP from PA is challenging and difficult to achieve through regular crystallization approaches. In this regard, we report four new salts of PAP with salicylic acid (SA), oxalic acid (OX), l-tartaric acid (TA), and (1S)-(+)-10-camphorsulfonic … Show more

“…The complete conversion of 4-aminophenol is vital, as any unconverted 4aminophenol is difficult to separate from paracetamol and will result in discolored and impure product. 16 With this modification, paracetamol was obtained in 96% yield at a concentration of 0.5 M, with 4% formation of 4-acetamidophenyl acetate [7], which was greatly reduced from the previous 14% formation.…”
