Optimizing the chitosan-cellulose based drug delivery system for controlling the ciprofloxacin release versus organic/inorganic crosslinker, characterization and kinetic study

Herein, interpenetrating network (IPN) hydrogels were successfully prepared for the first time through integrating two cross‐linking reactions in water in one pot, including amine‐anhydride reaction and subsequent photopolymerization. First, the polymer containing anhydride groups was prepared via free radical copolymerization of maleic anhydride and acrylamide. Second, the as‐prepared polymer was dissolved in water with polyacrylamine hydrochloride, N‐isopropylacrylamide, polyethylene glycol dimethacrylate, 2‐acrylamido‐2‐methylpropanesulfonic acid, and photoinitiator to obtain a homogeneous transparent solution. Third, the mixed solution was put in water bath of 50°C for 1 h to obtain the first network via the amine‐anhydride click reaction. Afterward, the second network was formed by photopolymerization. Finally, the swelling and drug release behavior of the hydrogels were studied with isoniazid as a model drug. The results show the IPN hydrogel possesses pH and temperature sensitivity and their controlled‐release behavior can be tuned via adjusting pH and temperature. In view of the advantages of amine‐anhydride reaction and photopolymerization, the strategy described here is of significance for the preparation of IPN hydrogel.

Cellulosic Polymers for Enhancing Drug Bioavailability in Ocular Drug Delivery Systems

One of the major impediments to drug development is low aqueous solubility and thus poor bioavailability, which leads to insufficient clinical utility. Around 70–80% of drugs in the discovery pipeline are suffering from poor aqueous solubility and poor bioavailability, which is a major challenge when one has to develop an ocular drug delivery system. The outer lipid layer, pre-corneal, dynamic, and static ocular barriers limit drug availability to the targeted ocular tissues. Biopharmaceutical Classification System (BCS) class II drugs with adequate permeability and limited or no aqueous solubility have been extensively studied for various polymer-based solubility enhancement approaches. The hydrophilic nature of cellulosic polymers and their tunable properties make them the polymers of choice in various solubility-enhancement techniques. This review focuses on various cellulose derivatives, specifically, their role, current status and novel modified cellulosic polymers for enhancing the bioavailability of BCS class II drugs in ocular drug delivery systems.

The valorization of industrial by-products is a green approach. This work investigates the valorization of kraft liquor from paper pulping as a precursor in the production of lignin-based nanocomposites to control the release of monoammonium phosphate (MAP) fertilizer. These nanocomposites were prepared from the stoichiometry of lignin nanoparticles (LNPs) and chitosan (Cs). The lignin nanoparticles were synthesized by solvent exchange and dialysis using two polar solvents: tetrahydrofuran (THF) and ethylene glycol (EG). These LNPs were characterized by transmission electron microscopy (TEM), X-ray diffraction (XRD), Fourier transform infrared (FTIR) spectroscopy, and textural techniques. The TEM images showed that the shapes of LNPs based on THF (LNPT) and LNPs based on EG (LNPE) were spherical, with an average particle size of 300 ± 40 nm and 23 ± 6.5 nm, respectively. Textural characteristics and FTIR revealed no change in the chemical nature of the two synthesized LNPs, and their Brunauer, Emmett and Teller (BET) surface area and average pore diameter ranged from 0.78 to 1.58 m 2 g −1 , and 4.069 to 5.20 nm, respectively. Swelling, functional groups, and the thermal, morphology, and MAP release characteristics were investigated for unloaded and loaded chitosan-LNP nanocomposites (Cs-LNPs). The water-swelling capacity of the nanocomposite gel was about 200%. The data indicated that the Cs-LNP gels can be used as controlled release systems for releasing MAP fertilizer, with maximum releasing value observed for chitosan-LNPT gel (65%). The kinetic of MAP release from the investigated nanocomposites was followed by the Ritger-Peppas model with Fickian and quasi-Fickian diffusion mechanisms.
