Abstract: The non-viral delivery of small RNA molecules like siRNAs still poses a major bottleneck for their successful application in vivo. This is particularly true with regard to crossing physiological barriers upon systemic administration. We have previously established polyethylenimine (PEI)-based complexes for therapeutic RNA formulation. These nanoplexes mediate full RNA protection against nucleolytic degradation, delivery to target tissues as well as cellular uptake, intracellular release and therapeutic efficac… Show more

“… 12 In contrast, PEI F25-LMW complexation likely facilitated entry of siRNA into brain tissue. This finding is also in line with recent studies in ex vivo tissue slice models demonstrating the penetration of PEI/siRNA complexes into solid tissues like tumors 39 , 40 and suggests that significant siRNA distribution could be achieved in the human brain, thus supporting further development of our approach. Most neurodegenerative diseases affect large brain regions or spread across the entire CNS.…”

“…Using explant cultures, anticancer drug screening has been demonstrated in rectal cancer liver metastases, which could be replicated in rodent xenograft models (Zhang et al, 2020). Tumor slices have also been utilized to test nanoparticle-based drug delivery of oligonucleotides to ovarian cancer and gliomas, and non-small cell lung cancer (Dong et al, 2011;Ewe et al, 2017).…”

“…Notably, even in the case of the smallest 2 kDa PEI, which is otherwise biologically inactive, tyrosine modification achieved knockdown efficacies of 50%. In addition, the colloidal stability of the tyrosine-modified PEI complexes upon incubation with serum was markedly improved [50][51][52]. More recently, we extended this strategy towards the modification of linear PEIs.…”
