Optimization of Process Conditions for the Production of Holocellulase by a Bacillus Species Isolated from Nahoon Beach Sediments

Abstract: Production of holocellulase by a newly isolated marine Bacillus species via submerged fermentation technique at varying cultivation conditions was investigated. Enzyme production was optimized by altering one variable per time. Moreover effect of lignocellulosic saccharides in inducing cellulase and xylanase production was also investigated. The 16S ribosomal Deoxyribonucleic Acid (16S rDNA) gene sequence analysis exhibited 98% sequence similarity of the isolate with other Bacillus species in the gene bank and… Show more

“…This indicates that production of peroxidase by the bacteria was growth associated. The decrease observed in the level of peroxidase produced by the bacteria after the optimal period of incubation may be as a result of denaturation or proteolysis (Fatokun et al., 2017). This finding contradicts a number of similar studies where the maximum peroxidase was produced at 72 h of incubation (Nour El-Dein et al., 2014; Falade et al., 2019b).…”

“…The levels of factors were selected by taking into account the operating experimental limits, and the previous studies. [34][35][36] The domain and matrix characteristics of Plackett-Burman experimental design used for the optimization of cellulase production were presented in Tables 1&2, respectively. As shown in Table 2, the experimental and predicted values of CMCase activity match quite well.…”

“…Thus, any alteration of cultivation temperature above or below the optimum temperature could lead to alteration in xylanase protein structure and change the three-dimensional shape or xylanase denaturation consequently altering its catalytic activities or its ability to bind substrates [30]. Resulting change in the reaction and lead to reduction in cellular metabolic activities rate, with subsequent inhibition in protein synthesis and cell growth [31].…”
