Optimization of isolation and cultivation of bacterial endophytes through addition of plant extract to nutrient media

Abstract: Many endophytes have beneficial effects on plants and can be exploited in biotechnological applications. Studies hypothesize that only 0.001–1% of all plant-associated bacteria are cultivable. Moreover, even after successful isolations, many endophytic bacteria often show reduced regrowth capacity. This research aimed to optimize isolation processes and culturing these bacteria afterwards. We compared several minimal and complex media in a screening. Beside the media themselves, two gelling agents and adding p… Show more

“…Finally, Chu10 and Bold's basal general media (Andersen, 2005) were selected for cyanobacteria isolation. In addition, Nutrient Agar and Tryptic Soy Agar (TSA) (Eevers et al, 2015) general media were selected for bacteria isolation. The soil bacteria were cultured and isolated according to standard protocols 6 described by Benson (2002).…”

“…From the resulting suspension, a series of 10‐fold dilutions down to 10 −4 were prepared. Aliquots (100 µl) of each dilution were spread in triplicates on M9 minimal medium, Gause’s synthetic agar (Zhao, Xu, & Jiang, ), Chitin medium (Lingappa & Lockwood, ), Tap Water Yeast Extract agar (El‐Shatoury, ), Humic acid‐vitamin (HV) agar (Hayakawa & Nonomura, ), Glycerol–asparagine agar (Pridham & Lyons, ) and 869 medium (Eevers et al, ). All media were supplemented with benomyl (50 µg/ml) and cycloheximide (50 µg/ml).…”
