Optical tweezer calibration by using a part of the intensity distribution of a trapped particle

Abstract: In this paper, we demonstrated that an optical tweezer setup can be calibrated by using a part of the symmetric intensity distribution of the trapped particle in digital video microscopy. First, we modified the radial symmetry center method, which was a recently proposed position detection algorithm. This algorithm uses gradient vectors of the particle intensity distribution, which allows us to use a part of the symmetric intensity distribution in the calculation of the particle center. We applied the modified… Show more

“…The force constant obtained with the 3 µm PS beads is about 7–8 pN/µm. This is on the lower end of the range of the force constants reached on different devices [ 3 , 7 , 11 , 39 , 40 , 41 , 42 ], but in some cases even smaller values were used [ 36 , 43 ]. Our device can, thus, be used for exerting the forces in the range of units, at most low tens, of pN/µm.…”

“…For CM, the accuracy on threshold effect is predictable as higher the threshold values will decrease the size of the active area. For the case of RSM, it is difficult to predict about the threshold effect due to the fact that the application of RSM with a threshold was performed first in [15,16] and it never be tested before. This makes the motivation of this work to investigate the threshold effect on the accuracy of the particle position using RSM and comparing the results with CM.…”

“…Here, we focus on to use a fraction of the particle intensity distribution for radially symmetric distribution. The gradient vectors allow us to apply it as shown in [24] and it provides an advantage to solve the overlapping problem in the particles images. If the particle intensity distribution covers only a several ten pixels area, using a part of the distribution can be useless.…”
