Opportunities and challenges related to sperm cryopreservation in Atlantic salmon gene banks

Abstract: Atlantic salmon are facing population declines and loss of productivity within populations due to anthropogenic impact factors and reduced survival at sea. Biobanking is an increasingly used tool to conserve the genetic integrity and diversity of populations threatened by extirpation. The aim of the current article is to discuss the opportunities and challenges that increased use of cryopreservation brings to biobanking activities, using the Norwegian Gene Bank (NGB) for Atlantic salmon as a model system. “…It is therefore pointless to invest in major biobanking projects that rely entirely on the use of cryopreserved, but untested, sperm samples. However, it is worth mentioning that the IUCN Species Survival Commission recently established an Animal Biobanking Specialist Group in 2022, thus endorsing the value that is currently being placed on biobanking as a further resource for species recovery programs (https://www.iucn.org/our-union/commissions/group/iucn-ssc-animalbiobanking-conservation-specialist-group) Agricultural and fisheries biobanks that focus on commercially significant species and breeds (Blackburn, 2018;Boe et al, 2021), as well as those focused on basic sciences and that store particular genetic lines of mice (Kaneko and Serikawa, 2012;Landel, 2005) and small fishes (Tiersch et al, 2011;Yang and Tiersch, 2009) ensure that their stocks of spermatozoa are reliably fertile. This level of assurance is not currently possible with most wild species, and it is for that reason that I regard only the Black-footed ferret and Giant panda projects as possessing well-established sperm biobanks.…”

“…The cryopreservation of sperm represents a multifaceted technique offering a plethora of advantages in fisheries management. These include the conservation of genetic diversity within fish populations, the preservation of gametes sourced from elite breeding individuals, facilitation of genetic material transport, and the provision of a reliable sperm reservoir independent of broodstock maturation synchronization (Judycka et al, 2019a;Bøe et al, 2021). However, despite considerable strides in standardizing cryopreservation protocols for fish semen, the practical application of cryopreserved sperm faces a myriad of challenges outweighing opportunities.…”

“…Because Yaqui Catfish are long-lived and iteroparous, broodstock that produce few surviving offspring can be respawned to retain their genetic variation in introduced populations. Cryopreservation of sperm can extend the reproductive life of males beyond their natural life span and can be used to slow the loss of genetic variation over time (Boe et al 2021).…”
