Novel Polymeric Nanocarriers Reduced Zinc and Doxycycline Toxicity in the Nematode Caenorhabditis elegans

Abstract: The objective was to evaluate the toxicity of zinc- and doxycycline-loaded polymeric nanoparticles (NPs) using Caenorhabditis elegans as a model organism. These NPs are composed of ethylene glycol dimethacrylate, 2-hydroxyethyl methacrylate and methacrylic acid. NPs were loaded with doxycycline (D-NPs) and zinc (Zn-NPs) by chemical adsorption, and loading efficacy was demonstrated. Worm death rate in a concentration-response curve basis was calculated for lethality. Metabolism was evaluated through pharyngeal … Show more

“…It has been previously reported that tetracyclines promote the proliferation of osteoblasts [ 4 ]; but, the underlying mechanism is not completely understood. Two different events may explain this fact: (i) osteoblasts’ proliferation is usually enhanced in the presence of antioxidants, as reactive oxidant species diminish osteoblasts’ reproduction [ 31 ], and doxycycline presents a potent antioxidant effect [ 32 , 33 ]. (ii) Doxycycline possesses calcium chelating ability that may facilitate calcium deposit on membranes [ 7 ].…”

“…M2 counts, higher in samples treated with both, Zn-HOOC-Si-Membranes and Dox-HOOC-Si-Membranes, have been shown to promote angiogenesis and vascular/matrix remodeling [48], which corresponded with the highest vasculature attained among groups in the present research (Tables 5 and 6). Additionally, M2 cells mainly participate in tissue remodeling and immunoregulation processes [12], decreased phagocytic capability [11], and strongly reduced the oxidative stress damage after using zinc and doxycycline [49]. The increase in the number of M2 macrophages leads to an immunological response shift through the production of anti-inflammatory cytokines and inhibition of reactive oxygen species [20].…”

“…We have established that industrial manufacturing procedures usually confer toxic properties on nanoparticles. The risk of exposure to these toxic components, however, depends on the likelihood and the extent of their exposure to living cells [120,121]. Thus, the in vitro and in vivo studies of toxicity assist to ascertain their toxic potential.…”
