Novel nanoimaging approach: Antibodious polymeric nanolabel for intracellular alpha‐fetoprotein targeted monitoring

Abstract: This study describes preparation and use of novel labeled and antibodious polymeric nanolabels (anti-alpha fetoprotein cross-linked nanolabels) as an immunogenic and semisynthetic nanolabel with potential prognostic and therapeutic roles for hepatoma cancer. Specificity, uptake, and binding efficiencies of the nanolabel have been examined in a human hepatosarcoma cell line HepG2, a human colorectal cell line DLD-1, and a mouse myoblast cell line C2. Labeling of the cells has been performed by treating live and… Show more

“…Those ruthenium‐based amino acid cross‐linkers provide a different cross‐linking procedure for biomolecules, which is based on the electron delocalization via ruthenium and the amino acids such as tyrosine, tryptophan, and so forth in the structure. After the cross‐linking of biomolecules with ANADOLUCA monomers at room temperature, under nitrogen atmosphere at daylight, the cross‐linked biomaterial gains stability for different reaction pH and temperature conditions 30–35 . The combination of the ANADOLUCA agent and MoSe 2 nanoparticles acts as the active site of the metalloenzyme FDH in this study, by the introduction of Mo, Se, and Cys moieties into the system.…”

“…The activation of the RIfS sensor surface was functionalized using the carbodiimide method with 70 µL, 1:1 NHS/EDC mixture in deionized water [28,29,34]. And for RIfS measurements under continuous ow was used degassed and ltered buffer (pH: 7.4, 10 mM phosphate buffer containing 0.5 % SDS) at a ow rate of 5 µLmin − 1 .…”

“…AmiNoAcid (monomer) Decorated and Light Underpinning Conjugation Approach (ANADOLUCA) provides a new effective perspective in complex heterogeneous processes, microemulsion polymerization, using photosensitive cross-linking strategies. According to the ANADOLUCA method, nanostructured proteins are synthesized through photo-electron transfer using ruthenium-based amino acid monomers in microemulsion polymerization media [27][28][29][30].…”

“…Size of synthesized nano enzyme could be determined by transmission electron microscopy (TEM), zeta‐sizer, and the molecular weight of the synthesized nano RuBisCO enzyme particles was determined by gel permanent chromatography (GPC). The enzymatic activity of the synthesized nano enzyme could be investigated using the same activity determination method with the free enzyme [21‐27].…”
