No evidence for a direct role of HLA-B27 in pathological bone formation in axial SpA

Abstract: ObjectiveThe strong genetic association between HLA-B27 and ankylosing spondylitis has been known for over 40 years. HLA-B27 positivity is possibly associated with severity of ankylosis. We studied the in vitro and in vivo impact of HLA-B27 in models of chondrogenesis and osteogenesis.MethodsDifferent in vitro differentiation systems were used to mimic endochondral and direct bone formation. ATDC5 cells and primary human periosteum-derived cells (hPDCs) were transduced with lentiviral vectors expressing HLA-B2… Show more

“…Contrary to the previously discussed literature, a study argues against a direct role of HLA-B27 in mediating new bone formation in axial SpA ( 106 ). HLA-B27 overexpression in various in vitro mouse and human differentiation systems mimicking endochondral ossification did not result in differences in bone formation when compared to the HLA-B7 overexpression controls.…”

“…ATDC5 cells can differentiate into chondrocytes. In addition, ATDC5 cells are widely used in the study of AS ( 23 , 24 ). LPS is commonly used as an inducer of inflammatory responses, stimulating cells to induce inflammatory responses and produce proinflammatory cytokines ( 25 ).…”

“…In SpA, inflammation of the sacroiliac and intervertebral joints is associated with both erosive bone changes and new bone formation at the entheses. HLA-B27 does not directly affect bone formation in transgenic mice or osteoblast differentiation by mouse or human bone precursor cells in vitro 80 ; rather, epigenetic and gene expression analyses suggest that new bone formation occurs in SpA owing to HLA-B27-driven inflammation 103,104 . The mechanistic hypotheses proposed to account for these findings are depicted in Fig.…”
