“…drugs, like glycerol trinitrate, or their precursors, like L-arginine, may play a role in helping to prevent preeclampsia through their action in relaxing blood vessel walls. [2] Serum nitrite levels were higher in preeclamptic women (34.11±14 ¼mol/l, P=0.04), lower in chronic hypertensive women (19.56±6.46 ¼mol/l, P=0.04) and similar in women with gestational hypertension (26.97±9.44 ¼mol/l) in comparison to the control group (25.37±7.24 ¼mol/l). [3] Circulating levels of nitrite are decreased in patients with preeclampsia.…”

“…Markers of oxidative stress are present in the placenta and maternal circulation of patients with pre-eclampsia (Raijmakers, Dechend & Poston, 2004) and we have previously shown reduced BH4 in placental extravascular vesicles from women with hypertensive pregnancies (Chuaiphichai et al, 2021). Yet in a landmark clinical trial the antioxidant vitamins C and E failed to prevent the development of pre-eclampsia in high risk pregnancies (Poston, Briley, Seed, Kelly & Shennan, 2006), and simple strategies attempting to restore or augment NO with NO donors in pre-eclampsia have been disappointing (Meher & Duley, 2007). One possible explanation for these results is the failure to specifically target eNOS uncoupling and consequent altered NO/ROS signalling.…”

“…Oral L-arginine supplementation was investigated by our group; the intervention did not alter blood pressure in preeclampsia (32). A recent review found that evidence is still insufficient to draw reliable conclusions as to whether L-arginine can prevent preeclampsia or its complications (33). Interestingly, a study involving dietary supplementation with 3 g/d L-arginine found no difference in biochemical markers, but supplementation significantly decreased maternal blood pressure and improved fetal outcome (34,35).…”
