Neuropeptides in sensory perikarya projecting to the rat ovary

Abstract: Afferent perikarya in dorsal root ganglia (DRG) at the T13 and L1 segmental levels projecting to the rat ovary were identified by utilizing the fluorescent retrograde tracer true blue (TB). Subsequently, TB-labeled ovarian afferent perikarya in DRG were examined for vasoactive intestinal polypeptide (VIP), substance P (SP), cholecystokinin-8 (CCK-8), neuropeptide Y (NPY), and somatostatin (SOM) immunoreactivity and for the presence of fluoride-resistant acid phosphatase (FRAP) enzyme activity. Of the ovarian a… Show more

“…1B and 2A), is suggestive of neuronal metaplasia. Although the presence of cholecystokinin-positive nerve fibers has been reported in the interstitium of the normal ovary (32,33), these are thought to originate from neurons situated in the dorsal root ganglia rather than an endogenous ovarian neurosecretory cell population, which is supported by our inability to detect Cck mRNA in normal ovaries (Fig. 1B).…”

“…Reverse transcription-PCR (RT-PCR) was done using the SuperScript One-Step RT-PCR System with Platinum Taq kit (Invitrogen, Carlsbad, CA), and %100 ng samples of ovarian and GCT total RNA that had been isolated as described above. Reactions were formulated as directed by the manufacturer, except that 0.625 ACi [a- 32 P]dCTP (specific activity, 3,000 Ci/mmol; MP Biomedicals, Irvine, CA) was added to each reaction to generate quantifiable radioactive signal and to increase assay sensitivity. Oligonucleotides used are detailed in Table 1, except those for ribosomal protein L19 (Rpl19), which were as described previously (ref.…”

“…We feel this is unlikely, however, since none of the other neuropeptides (CCK-8, substance P, VIP,) we have previously examined with dual labeling approaches in dorsal root ganglia have ever shown similar relationships to cells projecting to the ovary (McNeill and Burden, 1987).…”

“…An additional tissue used to control for the specificity of TB-labeling was the right T-13 dorsal root ganglia (DRG). Since retrograde labeling from the rat ovary is ipsilateral in DRG (Inyama et al, 1986;McNeill and Burden, 1987), any labeling in the opposite side at T-13 would indicate a non-specific, bloodborne label and provide a basis for rejection of the animal from the study. Both control tissues were sectioned and processed for visualization of CR-ir cells.…”

“…Several neurotransmitters and the enzymes participating in their synthesis or degradation (such as noradrenaline (NA), dopamine (DA), serotonin, acetyl-cholinesterase, gamma aminobutiric acid (GABA), VIP, SP, neuropeptide Y (NPY), cholecystokinin-8, the calcitonin gene-related peptide (CGRP), and corticotrophin releasing hormone (CRH)) are present in the fibbers innervating the internal theca of the follicles, the interstitial gland and the ovarian vasculature [14][15][16][17][18][19][20].…”
