Abstract: The development of neuronal circuitry required for cognition, complex motor behaviors, and sensory integration requires myelination. The role of glial cells such as astrocytes and microglia in shaping synapses and circuits have been covered in other reviews in this journal and elsewhere. This review summarizes the role of another glial cell type, oligodendrocytes, in shaping synapse formation, neuronal circuit development, and myelination in both normal development and in demyelinating disease. Oligodendrocyte… Show more

“…Hence, our study proposes an alternative non-cell-autonomous mechanism where OPC malfunction suppresses excitatory synaptogenesis thus contributing to pathogenesis of schizophrenia. Both excitatory and inhibitory synapses also connect neurons to OPCs [ 67 , 68 ], it is tempting to examine whether the neuron-OPC synapses are also disrupted in schizophrenia in future studies.…”

“…Preterm infants have structurally and functionally immature brains because major components of brain development including oligodendrocyte maturation, myelination, and synapse formation occur in the latter stages of gestation ( Jakovcevski et al, 2009 ; Stiles and Jernigan, 2010 ; Moura et al, 2022 ). Adequate brain development requires increased total mitochondrial capacity and higher ATP levels to meet the significantly increased rate of energy consumption ( Erecinska et al, 2004 ; Ten, 2017 ).…”

“…On top of this, OPCs were shown to form direct contacts with neuronal synapses, to participate in synaptic engulfment, and to engage in axon pruning during development and adulthood, thus contributing to circuit formation and modulation in mice ( Bergles et al, 2000 ; Auguste et al, 2022 ; Buchanan et al, 2022 ). Neuronal activity conveyed through these synapses relays information to OPCs that contributes to determining their proliferation, migration and differentiation, as well as oligodendrocyte survival, and myelination as a whole ( Mitew et al, 2018 ; Moura et al, 2022 ).…”
