Abstract: The endosomal sorting complexes required for transport (ESCRT) machinery functions in HIV-1 budding, cytokinesis, multivesicular body biogenesis, and other pathways, in the course of which it interacts with concave membrane necks and bud rims. To test the role of membrane shape in regulating ESCRT assembly, we nanofabricated templates for invaginated supported lipid bilayers. The assembly of the core ESCRT-III subunit CHMP4B/Snf7 is preferentially nucleated in the resulting 100-nm-deep membrane concavities. ES… Show more

“…The mechanisms of MV biogenesis still require further study. One accepted mechanism mediated by ESCRT machinery (Figure 4), which is similar to the classic mechanism of Exos biogenesis, showed that MVs could be formed via the recruitment of ESCRT‐III proteins 74, 75, 76. In addition to the ESCRT‐dependent mechanism, it has been hypothesized that there is also an ESCRT‐independent mechanism of MV biogenesis.…”

“…Nucleation is defined as the kinetic step in which an initial seed unit is assembled that once formed, can readily grow into a larger structure. In vitro analysis using negatively curved supported lipid bilayers created by nanofabrication showed that CHMP4B nucleates much faster on negatively curved membranes 69 . CHMP4B nucleation is also accelerated by the presence of ESCRT-II and CHMP6 69 .…”

CHMP4B nucleation is also accelerated by the presence of ESCRT-II and CHMP6 69 . In contrast, CHMP4B polymer growth is independent of both membrane curvature and upstream activators.…”

“…ESCRT-III filament formation can be promoted by membrane curvature (Lee et al 2015); by binding partners; and by modifications that favor the open state [or removal of modifications that restrict assembly (Crespo-Yàñez et al 2018)], concentrate the subunits, and/or properly orient subunits on a target membrane. The two best characterized nucleators of ESCRT-III polymerization, ALIX and ESCRT-II, appear to employ these principles, although their detailed mechanisms are not yet fully understood.…”
