Natural and Semisynthetic Pentacyclic Triterpenes for Chronic Myeloid Leukemia Therapy: Reality, Challenges and Perspectives

Abstract: Chronic myeloid leukemia (CML) is a neoplasm characterized by BCR‐ABL1, an oncoprotein with vital role in leukemogenesis. Its inhibition by tyrosine kinase inhibitors represents the main choice of treatment. However, therapeutic failure is worrying given the lack of pharmacological options. Pentacyclic triterpenes are phytochemicals with outstanding antitumoral properties and have also been explored as a basis for the design of potential leads. In this review, we have gathered and discuss data regarding both n… Show more

“…The semisynthetic modifications of pentacyclic triterpenes provide a broad variety of functionalities, from increasing potency and/or selectivity to optimizing pharmacokinetic parameters [ 73 ]. One of the several nitrogen heterocyclic compounds present in nature, the indole ring is often present in plant hormones such as tryptophan and auxins, serotonin or 5-hydroxytryptamine, and melatonin [ 74 ].…”

“…[74,75] Some steroids and terpenes could exert antileukemic activity through various pathways including the activation of mitochondrial apoptosis, whereas some of them are deemed as sensitizing leukemia cells to the chemotherapeutic effects of many cytotoxic agents, primarily by inhibiting efflux pumps, thus reducing leukemia resistance. [76,77] Accordingly, a combination of 1,2,3-triazole with steroid/terpene is a promising strategy to develop novel antileukemic chemotherapeutics.…”

“…These triterpenoids’ properties, along with previous reports on pentacyclic triterpenes’ action against leukemia cells [ 7 , 12 ], suggested to us the need to assess their effects in chronic and acute myeloid leukemia cells. The present study objective was to evaluate the effect of these metabolites against the K562 and HL60 myeloid leukemia cell lines.…”
