Multilocus sequence typing and virulence factors analysis of Escherichia coli O157 strains in China

Abstract: Escherichia coli O157:H7, an important food-borne pathogen, has become a major public health concern worldwide. The aim of this study was to investigate the molecular epidemiologic feature of E. coli O157:H7 strains in China. 105 E. coli O157:H7 isolates were collected from various hosts and places over 9 years. A multilocus sequence typing scheme (MLST) was applied for bacteria genotyping and polymerase chain reaction (PCR) was used for virulence factor identification. Seven new MLST sequence types (STs), nam… Show more

“…A multiplex PCR procedure was used to detect the stx1 and stx2 genes ( 30 ), and a single PCR procedure was used to detect the eae ( 31 ), hly ( 32 ), and tccp ( 14 ) genes. The primers, conditions, and references cited are listed in Table 1 .…”

“…The LEE region also encodes intimin, an outer membrane protein adhesin (Eae) that mediates the intimate attachment of bacteria to the host epithelial cell surface ( 13 ). In addition, Tir cytoskeleton-coupling protein (TccP) stimulates actin polymerization during the formation of A/E lesion ( 14 ). The large plasmid pO157 carries genes coding for type II secretion systems, such as hemolysin (Hly) and ToxB.…”

“…Nucleotide sequences of E. coli O157:H7 str. Sakai (NCBI Reference Sequence: BA000007, ST11) [29] were obtained from the NCBI repository and included in subsequent analyses for comparison. To analyse the distribution of STs in freerange mink farms, a minimum spanning tree was generated using BioNumerics software, version 7.6 (Applied Maths, Kortrijk, Belgium).…”

“…Samples were homogenized and seeded on MacConkey plates (Oxoid, Basingstoke, UK) or in McConkey agar containing disks of ceftazidime (30 μg) at one end of the plate and incubated at 37°C for 24 h. Then, isolated colonies compatible with E. coli growing within the ceftazidime-treated halos or without antibiotic pressure were selected and confirmed by polymerase chain reaction (PCR) amplifying the E. coli -specific uidA gene [ 21 ].…”
