Abstract: Mucosal-associated invariant T (MAIT) cells are unique innate-like T cells that bridge innate and adaptive immunity. They are activated by conserved bacterial ligands derived from vitamin B biosynthesis and have important roles in defence against bacterial and viral infections. However, they can also have various deleterious and protective functions in autoimmune, inflammatory and metabolic diseases. MAIT cell involvement in a large spectrum of pathological conditions makes them attractive targets for potentia… Show more

“…Here we showed that MAIT17 cells are a major contributors of IL-17 in children with CAP, particularly in early childhood. However, while involvement of MAIT cells in anti-microbial immunity and pathogenesis of human diseases is implicated 11 , direct evidence that MAIT cells mediate immune defense and immunopathology in human diseases including CAP is still lacking. Of note, scRNAseq analysis showed that MAIT17 from CAP patient BAL expressed GRP65.…”

“…In summary, MAIT cells are increasingly appreciated as participants in immune protection and the pathogenesis of human infectious and non-infectious diseases 11 . IL-17 production is a characteristic and yet enigmatic feature of MAIT cells.…”

“…This TCR specificity restricts MAIT cells to antigens presented by the non-polymorphic MHC class-I related (MR1) molecule, which is highly conserved across mammals (5)(6)(7)(8). The pathogenic and protective roles of MAIT cells in autoimmune and inflammatory disease have been extensively studied (9)(10)(11)(12) but their role in cancer is unclear. Although there are currently no reports of tumor antigens presented by MR1 to MAIT cells (13)(14)(15)(16), MAIT cells upregulate granzyme B and perforin following activation (5,17,18) and have the capacity to kill tumor cells pulsed with the MAIT cell antigen 5-(2oxopropylideneamino)-6-D-ribitylaminouracil (5-OP-RU) (19), suggesting that MAIT cells are capable of eliciting direct anti-tumor immune responses.…”

“…1 MAIT cells, in particular IL-17-producing subsets, have been implicated in numerous chronic inflammatory diseases including rheumatoid arthritis, ankylosing spondylitis and psoriasis. 2 We have previously described alterations in MAIT cell cytokine profiles in obesity, including reduced IFNy production and elevated IL-17 production. 3 More recently, we have reported alterations in MAIT cell metabolism in patients with obesity that underpin the loss of IFNy production by MAIT cells.…”
