Morphogen gradients, positional information, and Xenopus: Interplay of theory and experiment

Abstract: The idea of morphogen gradients has long been an important one in developmental biology. Studies with amphibians and with Xenopus in particular have made significant contributions to demonstrating the existence, identity, and mechanisms of action of morphogens. Mesoderm induction and patterning by activin, nodals, bone morphogenetic proteins, and fibroblast growth factors have been analyzed thoroughly and reveal recurrent and combinatorial roles for these protein growth factor morphogens and their antagonists.… Show more

“…Differential Nodal activity, moreover, appears to lead to differential cell fates within the NO derivatives: initially, high levels appear to induce the formation of the AME and endoderm, whereas lower levels induce greater mesoderm development (reviewed in Osada and Wright, 1999;Joseph and Melton, 1998;Kiecker and Niehrs, 2001;Whitman, 2001;Green, 2002;Schier, 2003). These studies further serve to highlight the idea that the appropriate balance of NO-derivative cell fates (endoderm, mesoderm, and mesendoderm) must be achieved.…”

“…These include signalling from the Nieuwkoop Centre, the Spemann Centre, and the notochord (Green, 2002;Marti and Bovolenta, 2002;Ashe and Briscoe, 2006). Patterning of embryonic tissue by such morphogens is well established, but the mechanism by which such a patterning occurs is still obscure.…”

“…The organized operation of cell behaviors depends on temporal and spatial references settled by signaling centers, i.e., zones of polarizing activity (Freeman and Gurdon, 2002;Green, 2002). The isthmic organizer (IsO), a signaling center located at the midhindbrain (MHB) isthmus, regulates the patterning along the cephaliccaudal (cph-cd) axis of both the midand the hindbrain (Joyner et al, 2000;Olander et al, 2006).…”
