Abstract: Cytochrome c (Cyt c) has key roles in both mitochondrial electron transfer and apoptosis onset and is therefore likely undergoing a strong selective pressure against amino acid variation. “…To further investigate the role of the Arg38 sidechain in the peroxidase reaction, we expressed the R38W variant of cytochrome c. This variant was originally reported as a low frequency SNP (rs11548791, p.Arg39Trp, (38)). However it is now unclear if this is a real variant (39), and gnomAD v2.1.1 reports an alternative low frequency SNP (rs1047899818, p.Arg39Gln) (40). Nonetheless, as Trp has a larger van der Waals volume than Arg (0.79 nm 3 compared to 0.59 nm 3 ) and therefore might be expected to block access to substrate, we hypothesized that this variant would decrease the peroxidase activity of cytochrome c.
