Molecular and genetic characterization of the Ryadg locus on chromosome XI from Andigena potatoes conferring extreme resistance to potato virus Y

Abstract: Key messageWe have elucidated the Andigena origin of the potato Ryadg gene on chromosome XI of CIP breeding lines and developed two marker assays to facilitate its introgression in potato by marker-assisted selection.AbstractPotato virus Y (PVY) is causing yield and quality losses forcing farmers to renew periodically their seeds from clean stocks. Two loci for extreme resistance to PVY, one on chromosome XI and the other on XII, have been identified and used in breeding. The latter corresponds to a well-known… Show more

“…Emma genotyping as resistant using both markers yet is phenotypically classified as susceptible to PVY. Low association between RYSC3 marker alleles and Ry adg mediated resistance was also reported by Dalla Rizza et al [26] and Herrera et al [27]. To this effect, use of the RYSC3 marker has been replaced by alternative markers such as M45 and M6 [23,27].…”

To this effect, use of the RYSC3 marker has been replaced by alternative markers such as M45 and M6 [23,27]. There is currently no formal evidence in the literature to suggest that the M6 marker incorrectly genotypes any samples, however, it has only recently been reported and has not been subjected to close scrutiny or evaluation.…”

“…Wild potato varieties and landraces are sources of PVY-specific NLR resistance genes that can be introgressed into commercial potato cultivars. Loci conferring extreme resistance to PVY have been mapped in Solanum chacoense (Ry chc ), S. tuberosum group Andigena (Ry adg ), and S. stoloniferum (Ry sto ; Herrera et al, 2018). Grech-Baran et al (2020) employed resistance enrichment sequencing (RenSeq) to isolate the gene conferring Ry sto -mediated extreme resistance from the commercial potato cultivar, Alicja.…”

“…Marker assisted selection can also screen at early stage major genes and Quantitative Trait Loci (QTL) with large effects (Gebhardt 2013;Sharma et al 2014). In recent years, markers flanking major genes and QTL were developed for resistance to viruses (Mihovilovich et al 2014;del Rosario et al 2018), tuber starch and yield (Schönhals et al 2016), and other important traits (Ramakrishnan et al 2015). If applied at the early clonal generation stage and multiplexed, marker-assisted selection can be cost-effective (Slater et al 2013).…”
