This study aimed to investigate the combined effects of two most potent probiotic bacteria Lactobacillus acidophilus and Lactobacillus plantarum on overall health and immune status of freshwater crayfish, marron under laboratory conditions. A total of 36 marron were distributed into six different tanks and two different feeding groups, control and probiotic-fed group. After acclimation, control group was fed with basal diet while probiotic group was fed 10 9 cfU/mL per kg of bacterial supplemented feed for 60 days. The results showed no significant differences in weight gain, however, probiotic feed significantly enhanced some hemolymph parameters and biochemical composition of tail muscle. Histology data revealed better hepatopancreas health and higher microvilli counts in the marron gut fed probiotic diet. The probiotic bacteria triggered significant shift of microbial communities at different taxa level, mostly those reported as beneficial for crayfish. The probiotic diet also enriched the metabolic functions and genes associated with innate immune response of crayfish. Further correlation analysis revealed significant association of some taxa with increased activity for hemolymph and immune genes. therefore, dietary Lactobacillus supplementation can modulate the overall health and immunity as well as gut microbial composition and interaction network between gut microbiota and immune system in crayfish. Aquaculture has become an important food sector for meeting the overall protein demand for growing population. The global consumption of crustaceans has increased greatly and especially those with live transport abilities are increasingly becoming popular 1-3. Marron (Cherax cainni) is one of the largest freshwater crayfish farmed in Western Australia (WA) that has high nutritive value and widespread consumer preferences 4,5. In addition, long distance live transportation ability of marron further increases its international demand, and thus has become an ideal crayfish species for commercial farming 6,7. However, the production of marron in WA has remained stagnant for a long time 7. Selection of proper diets and maintaining the optimum water quality are the two most crucial factors required for marron farming 5,7. Although no disease outbreaks have yet been reported, the interest and intend of intensification in marron farming can expose marron to possible crayfish pathogens including Vibrio, Aeromonas and Rhodobacter 5,6. In the past, several laboratory based trials have been conducted 3,6-9 , however, finding a suitable diet and identifying the beneficial bacteria that potentially can influence the growth and immune performance of marron has still remained unknown. The employment of traditional bait and medicament in freshwater aquaculture causes the frequent occurrence of environmental pollution and disease. Effluent collected after wastewater treatment with Rhodopseudomonas sphaeroides could be reutilized for microbial feed, medicament, and aquaculture water to culture Common Carp Cyprinus carpio. Therefore, a novel integrated system of wastewater treatment and the use of the effluent containing R. sphaeroides, which could improve yield, increase disease resistance, and enhance the quality of aquaculture water for Common Carp culture, was proposed and investigated. Common Carp can grow well in effluent containing R. sphaeroides. The survival rate, yield, and whole‐body composition of the fish raised in effluent containing R. sphaeroides were all increased compared with the control group. The biochemical (B vitamin) and other substances in the effluent with R. sphaeroides enhanced the activity of proteases, amylases, lipases, alkaline phosphatase, acid phosphatase, phagocytes, superoxide dismutase, and catalase by up‐regulating the expression of their associated genes. Theoretical analysis showed that biochemical molecules regulate the expression of these gene and enzyme activities by acting as a signal that stimulates the active center. Moreover, biochemicals present in R. sphaeroides enhanced the mTOR and NF‐kB signaling pathways and improved intestinal microbiota. Furthermore, R. sphaeroides inhibited Aeromonas hydrophila, increasing resistance against fish disease and promoting the growth of intestinal bacteria. Meanwhile, R. sphaeroides in the effluent also improved the aquaculture water quality. This technology would simultaneously save water in aquaculture, reduce water pollution and wastewater discharge, and increase the output and disease resistance of Common Carp.

This study aimed to investigate the effectiveness of synbiotic (prebiotic alginate and probiotic bacteria Micrococcus spp) on the immune responses, growth performance, and bacterial resistance of freshwater crayfish. The experimental diets were formulated in three levels of synbiotic: the probiotic bacteria Micrococcus spp+100 mg/L alginate (Syn100), Micrococcus spp+200 mg/L alginate (Syn200), Micrococcus spp+300 mg/L alginate (Syn300), each level was mixed with tested feed. The basal diet (without synbiotics) served as the control diet. During 40 days of rearing, immune responses observations were conducted every 10 days, while the crayfish weight were recorded on day 40. Furthermore, the challenge test was performed against the pathogenic bacteria of A. hydrophila and reared for a week. The result showed that synbiotics treatment of Syn300 and Syn200 could improve the immune response and increased the growth rate (P<0.05). Both were also heavier than the Syn100 treatment. The highest resistance to A. hydrophila pathogenic bacteria by demonstrating a higher survival rate was Syn300 and followed by Syn200 (70.00 ± 0% and 73.33 ± 5.77%, respectively), compared to Syn100 (56.67 ± 3.33%) and the control (33.33 ± 3.33%). The synbiotic Micrococcus spp. combined with alginate potentially induced immune responses, increased growth performance, and improved bacterial pathogens resistance, making it an ideal synbiotic to be developed. Keywords: synbiotic, prebiotic, probiotic, Sargassum sp., alginate, brown algae ABSTRAK Penelitian ini bertujuan untuk menguji efektivitas penggunaan sinbiotik (prebiotik alginat dan probiotik Micrococcus spp) terhadap respons imun, performa pertumbuhan dan resistensi lobster air tawar. Tiga perlakuan sinbiotik yaitu Micrococcus spp+100 mg/L alginat (Syn100), Micrococcus spp+200 mg/L alginat (Syn200), Micrococcus spp+300 mg/L alginat (Syn300), masing-masing dicampurkan ke dalam pakan dan diberikan ke lobster. Pakan tanpa campuran prebiotik digunakan sebagai pakan kontrol. Selama 40 hari pemeliharaan, pengamatan respon imun dilakukan setiap 10 hari dan pada hari ke-40 pemeliharaan dilakukan pengukuran bobot lobster. Selanjutnya, lobster di uji tantang dengan bakteri A. hydrophila dan dipelihara kembali selama satu minggu. Hasil penelitian menunjukkan bahwa pemberian pakan yang mengandung sinbiotik Syn300 dan Syn200 dapat meningkatkan respon imun dan pertumbuhan (P<0.05) lebih tinggi dibandingkan dengan perlakuan Syn100, sementara bobot tubuh terendah didapat pada perlakuan kontrol. Resistensi lobster tertinggi terhadap bakteri A. hydrophila yang ditunjukkan dengan sintasan tertinggi didapatkan pada perlakuan Syn300 (70.00±0.00%) dan Syn200 (73.33±5.77%) dibandingkan dengan perlakuan Syn100 (56.67±3.33%) dan kontrol (33.33±3.33%). Penelitian ini membuktikan bahwa sinbiotik Micrococcus spp. yang dikombinasikan dengan alginat dapat menginduksi respons imun, meningkatkan performa pertumbuhan ,dan resistensi terhadap patogen sehingga merupakan sinbiotik yang ideal untuk dikembangkan.
