Abstract: Mitochondria are essential organelles in physiology and kidney diseases, because they produce cellular energy required to perform their function. During mitochondrial metabolism, reactive oxygen species (ROS) are produced. ROS function as secondary messengers, inducing redox-sensitive post-translational modifications (PTM) in proteins and activating or deactivating different cell signaling pathways. However, in kidney diseases, ROS overproduction causes oxidative stress (OS), inducing mitochondrial dysfunction… Show more

“…As mentioned above, mitochondrial metabolism is a source of ROS that directly modulates cell-signaling pathways, protein function through post-translational modifications, and influences cell survival, proliferation, and apoptosis [ 46 ]. However, increased ROS production results in oxidative stress that causes damage to lipids, proteins, and nucleic acids, eventually disrupting cellular homeostasis.…”

“…Upregulation of renal NOX4 plays an important role in several pathologies, like DKD, hypertensive nephropathy, and polycystic kidney disease by increasing ROS levels and mitochondrial damage [ 51 , 52 , 53 , 54 , 55 ]. It has been described that mitochondrial ROS and NOX-produced ROS can enhance damage and depolarization of the mitochondrial membrane potential [ 46 , 56 , 57 ]. The total absence of NOX4 has also been reported to be pathological since it results in fibrosis and oxidative stress [ 58 ].…”

“…In CKD models, NOX4 is the principal isoform activated due to Ang II upregulation [ 247 ]. NOX4 is highly expressed in cellular membranes like mitochondria and ER, and essentially produces H 2 O 2 through histidine groups in its E-loop that permits the rapid dismutation of O 2 •− to H 2 O 2 [ 248 , 249 ].…”

“…Oxidative stress has been reported in kidney disease, due to both antioxidant depletions as well as increased production of ROS (Daenen et al 2019;Irazabal and Torres 2020). The kidney is a highly metabolic organ, rich in oxidation reactions in mitochondria, which makes it vulnerable to damage caused by ROS (Aranda-Rivera et al 2021). Therefore, OS can accelerate kidney disease progression.…”
