MiR-425 expression profiling in acute myeloid leukemia might guide the treatment choice between allogeneic transplantation and chemotherapy

Abstract: BackgroundAcute myeloid leukemia (AML) is a highly heterogeneous disease. MicroRNAs function as important biomarkers in the clinical prognosis of AML.MethodsThis study identified miR-425 as a prognostic factor in AML by screening the TCGA dataset. A total of 162 patients with AML were enrolled for the study and divided into chemotherapy and allogeneic hematopoietic stem cell transplantation (allo-HSCT) groups.ResultsIn the chemotherapy group, patients with high miR-425 expression had significantly longer overa… Show more

“…Clear evidence suggests that miRNAs play a role in many aspects of AML development, including maintenance of hematopoietic stem cells, growth, proliferation, differentiation, and apoptosis . Moreover, changes in the expression of different types of microRNA have been correlated with prognosis in cases of AML . Gaining the ability to reverse these changes may have therapeutic implications and improve the outcome for these patients…”

“…16 Moreover, changes in the expression of different types of microRNA have been correlated with prognosis in cases of AML. 26 Gaining the ability to reverse these changes may have therapeutic implications and improve the outcome for these patients. 15 Among the different types of miRNAs, miR-204 has been the subject of numerous studies in solid cancers.…”

“…The encoded proteins fulfil a wide range of functions with signaling (7 genes), transcription (8 genes) and metabolism (10 genes) being the most prominent (Supplementary Table S9). Five of these genes have been studied in AML (see [16,18,19,20,21,22]).…”

“…High miR-425 level was associated with significantly longer OS and event-free survival (EFS) in non-transplant patients, but this association was not observed in post allo-HSCT patients. Instead, patients with downregulated miR-425 did better if they had allo-HSCT, suggesting that low miR-425 level might be an indication for transplant [79]. Overexpression of miR-99a predicted adverse prognosis in AML patients irrespective of transplant status, necessitating the investigation of novel alternative treatment in miR-99a overexpressors [80].…”
