Microsequence analysis of the N‐terminally blocked proteins immobilized on polyvinylidene difluoride membrane by Western blotting

Abstract: A technique has been developed for efficient deblocking and subsequent microsequencing of N-terminally blocked proteins immobilized on a polyvinylidene difluoride (PVDF) membrane at the picomole levels. In this technique, proteins were first separated by polyacrylamide gel electrophoresis and then transferred onto a PVDF membrane by Western blotting. The electroblotted proteins with N-terminal acetylserine or acetylthreonine could be deblocked on-membrane by treatment with trifluoroacetic acid vapor and sequen… Show more

“…Their N-terminus is blocked by acetylation. The N-terminal amino acid is acetyl-Ser, as in the other inamrnalian [-chains, shown by Edman degradation after the intact chain was subjected to prolonged exposure to triflouroacetic acid which partialy removes the acetyl group (Hirano et al, 1993).…”

“…Both the zeta-chains (c and [), of Hbs 3 and 4 were N-terminally blocked, a feature common to the embryonic [ chains of mammals. To determine the N-terminal sequence, the Hb4 5 chain was first subjected to mild acid hydrolysis according to Hirano et al (1993) and subsequently sequenced by Edman degradation. To determine further partial primary structure analysis of one of Table 1.…”

“…MALDI-TOF MS analysis of a tryptic digest of the protein indicated for the N-terminal peptide MDSQVR a value of m/z 777, which points to the presence of acetylmethionine. This residue is commonly found in the N-terminus of eukaryotic proteins together with N-acetyl Ala, Ser, Gly and Thr [17].…”

“…Application of four different deblocking strategies [9] did not solve the problem. Thus the aim of the present investigation was to characterize this potential allergen further and, in particular, to obtain sequence data which may highlight its function in the plants.…”
