MicroRNA-Mediated Gene Silencing in Plant Defense and Viral Counter-Defense

Abstract: MicroRNAs (miRNAs) are non-coding RNAs of approximately 20–24 nucleotides in length that serve as central regulators of eukaryotic gene expression by targeting mRNAs for cleavage or translational repression. In plants, miRNAs are associated with numerous regulatory pathways in growth and development processes, and defensive responses in plant–pathogen interactions. Recently, significant progress has been made in understanding miRNA-mediated gene silencing and how viruses counter this defense mechanism. Here, w… Show more

“…These molecules play an important role in posttranscriptional regulation by the inhibition of the expression of target genes by binding to mRNA [1]. Eukaryotic organisms, such as plants and animals, and some viruses express this type of molecules [2,3]. Lin-4 is the first miRNA that was identified in Caenorhabditis elegans, although later in the same organism were found 22-nt lin-4 and 21-nt let-7; currently, more than 18,226 miRNAs are reported [4].…”

“…miR395 is another miRNA that has an influence on PA biosynthesis [30]. Some miRNAs (U436803, U977315, U805963, U3938865, and U4351355) regulate fatty acid and flavonoid biosynthesis in Lonicera japonica [2,3]. The characterization in A. thaliana shows that miR858a targets MYB transcription factors that are involved in flavonoid biosynthesis, growth, and development.…”

“…For this significant discovery, Fire and Mello were awarded with the Nobel Prize in Physiology or Medicine in the year 2006. Since then, this phenomenon has been extensively applied to suppress gene function and has become a common tool for the functional analysis of the genome (Sharma et al, 2013;Younis et al, 2014;Mermigka et al, 2015;Guo et al, 2016;Liu et al, 2017;Carbonell and Darosa 2017;Hua et al, 2018). Some applications of gene silencing have been represented in the Table 1. Key Features of RNAi includes, silencing at post transcriptional level, double stranded nature of silencing molecule, presence of catalytic and amplification process, heritable and systemic silencing, extremely high specificity of the silencing process.…”

“…A full complementarity binding between target mRNA and miRNA sequence results in endonucleolytic cleavage and disruption. Contrary to this, partial target-site complementarity characteristically down-regulates target-gene expression by suppressing translation of target mRNA (Liu et al 2017). The present study identifies suitable sugarcane-encoded miRNAs to exhibit a stronger degree of target-site complementarities within the ORF1, ORF2 and ORF3 of SCBGAV.…”
