Micro-Optical Sectioning Tomography to Obtain a High-Resolution Atlas of the Mouse Brain

Abstract: The neuroanatomical architecture is considered to be the basis for understanding brain function and dysfunction. However, existing imaging tools have limitations for brainwide mapping of neural circuits at a mesoscale level. We developed a micro-optical sectioning tomography (MOST) system that can provide micrometer-scale tomography of a centimeter-sized whole mouse brain. Using MOST, we obtained a three-dimensional structural data set of a Golgi-stained whole mouse brain at the neurite level. The morphology a… Show more

“…A three-dimensional structural data set of a Golgi-stained whole brain at the neurite level has been obtained to explore connectional groups and functional groups in the basic structure and function of neural circuitry [2] . The dominant view that a saliency map is generated in the parietal cortex has been challenged because a bottomup saliency map is created in V1 [1] .…”

“…Thus far, providing an image at organelle-level resolution of a whole organ has remained a challenge. Recently, optical microscopy assisted by chemical clearing or mechanical sectioning of tissue has been proven as a powerful technique to overcome this dilemma [1][2][3] . However, this type of techniques requires lengthy special preparation of the tissue specimen, hindering their applicability in life sciences.…”
