Methanococcus thermolithotrophicus, a novel thermophilic lithotrophic methanogen

Abstract: Abstract. An autotrophic thermophilic motile coccoid methanogen was isolated from geothermally heated sea Sediments close to Naples, Italy. Growth occurs on H 2 /C0 2 and on formate between 30 and 70° C with an Optimum at 65° C. The optimal doubling time is only 55 min. The NaClconcentration ranges from 1.3% to 8.3% with an Optimum around 4%. By its G + C-content of 31.3 mol %, its subunit envelope, and by DNA-RNA hybridization the new isolate is clearly defined to be a member of the genus Methanococcus. We na… Show more

“…It is the first hydrogen-oxidizing methanogen growing between 0 and 12.5 YO NaCl, a range which is the widest reported to date for any hydrogenotrophic methanogen. To our knowledge, the upper NaCl limit for growth of hydrogenotrophic methanogenic archaea is 8-3 YO reported for Methanococcus thermolithotrophicus (Huber et al, 1982). Strain SEBR 4845T is an irregular coccus and has a G + C content of 55 mol YO.…”

“…Strain M7T differs from M. thermolithotrophicus strain SN-lT in its inability to use formate and in its temperature range and optimum for growth (Table 1) (Huber et al, 1982;Jones et al, 1989). It differs from M .…”

“…The source of 'Methanococcus aeolicus' has not been reported. Two strains of Methanococcus thermolithotrophicus were isolated from coastal geothermally heated sediments close to Naples (Italy) (Huber et al, 1982) and from reservoir water from a North Sea oilfield (Nilsen & Torsvik, 1996). The selenium-independent hyperthermophilic methanogen Methanococcus igneus was isolated from a shallow submarine hydrothermal vent (Kolbeinsey ridge, Iceland) (Burggraf et al, 1990).…”

“…Methanococcus infernus strain MET (DSM 1 1812T) and Methanococcus vulcanius M7T (DSM 1 2094T) were isolated in our laboratory (Jeanthon et al, 1998(Jeanthon et al, , 1999. Methanococcus jannaschii strain JAL-lT (DSM 266 lT), Methanococcus fervens strain AG86T (DSM 421 3T), Methanococcus igneus strain Kol 5T (DSM 5666T) and Methanococcus thermolithotrophicus strain SN-1 (DSM 2095T) (Huber et al, 1982; Jones et al, 1983;Zhao et al, 1988; Burggraf et al, 1990;Jeanthon et al, 1999) were obtained from the Deutsche Sammlung von Mikroorganismen und Zellkulturen (Braunschweig, Germany).Genomic DNA extraction and purification were carried by using the procedure described by Charbonnier & Forterre (1994). Amplified 16s rDNA was obtained from each strain by PCR with the following specific archaeal primers : forward 5' TTCCGGTTGATC-CYGCCGGA 3', reverse 5' GGTTACCTTGTT-ACGACTT 3', corresponding to the positions 2-2 1 and to 151&1492 of Escherichia coli 16s rRNA, respectively (Brosius et al, 1978).…”
