Metastasis-inducing S100A4 protein is associated with the disease activity of rheumatoid arthritis

Abstract: This is the first study to show that the levels of the S100A4 protein are correlated with RA disease activity. Furthermore, only the bioactive form, but not the total amount of S100A4, decreases after successful TNF-alpha blocking therapy in patients with RA. These data support an important role for the S100A4 multimer in the pathogenesis of RA.

“…Expression of S100A4 is enhanced in several chronic inflammatory conditions, and concentrations are correlated with disease activity (20). S100A4 induces the production of pro-inflammatory proteins such as TNF-a, IL1b and IL6 in mononuclear cells (38).…”

“…Of note, circulating levels of these proteins have been correlated to inflammation and increased risk of CVD (19,20,21,22). Due to the close connection between the S100 proteins and the receptor for advanced glycation endoproducts (RAGE), we also analysed RAGE in order to have more information on the inflammatory process in patients with PHPT.…”

“…18 This finding is consistent with the increased level of MMP-9 reported in tears of subjects with CCH. 7 In addition, Oslejsková et al 19 speculated that the S100A4 protein that is located at sites of inflammation, particularly at sites of joint destruction, might be implicated in the pathogenesis of chronic autoinflammatory diseases. It thus seems that the presence of conjunctival folds while occupying the lower tear meniscus leads to a delay in tear clearance, which in turn leads to an alteration in tear composition because delayed tear clearance increases the residency time of the contents of the tear film and may activate or potentiate ocular surface inflammation.…”

“…Our results showed, that in inflamed muscle, the S100A4 protein is produced mainly by mononuclear cells present in the inflammatory infiltrates, by endothelial cells and by regenerating muscle fibres [22]. Moreover, we have previously found increased circulating levels of S100A4 in patients with rheumatoid arthritis (RA) in comparison with control individuals and demonstrated a positive correlation between S100A4 and disease activity in RA [23]. …”

“…Serum S100A4 concentrations were measured by an ELISA kit according to the manufacturer’s protocol (CycLex Co, Ltd, Ina, Nagano, Japan) as demonstrated elsewhere [23]. The analysis was performed using the ELISA reader SUNRISE (Tecan, Salzburg, Austria) at a wavelength of 450 nm.…”
