Metabolic Plasticity and Epithelial-Mesenchymal Transition

Abstract: A major transcriptional and phenotypic reprogramming event during development is the establishment of the mesodermal layer from the ectoderm through epithelial-mesenchymal transition (EMT). EMT is employed in subsequent developmental events, and also in many physiological and pathological processes, such as the dissemination of cancer cells through metastasis, as a reversible transition between epithelial and mesenchymal states. The remarkable phenotypic remodeling accompanying these transitions is driven by c… Show more

“…As such, we characterized the EMT spectrum in depth, finding numerous enriched signaling, metabolic, and regulatory features throughout. Amongst these, in late-hybrid EMT states, we observed increased MYC activity and proliferation, as well as potential metabolic rewiring from OXPHOS to glycolysis, which has been implicated in both tumor invasiveness 49,50 and EMT 51,52 .…”

“…The epithelial-mesenchymal transition (EMT) is a cell biological process crucial for various aspects of tumor aggressivenesscancer metastasis , resistance against cell death (Huang et al, 2013), metabolic reprogramming (Thomson et al, 2019), refractory response to chemotherapy and radiotherapy (Kurrey et al, 2009), tumor-initiation potential (Jolly et al, 2014), and immune evasion (Tripathi et al, 2016;Terry et al, 2017)thus eventually affecting patient survival (Tan et al, 2014). EMT is a multidimensional, non-linear process that involves changes in a compendium of molecular and morphological traits, such as altered cell polarity, partial or complete loss of cell-cell adhesion, and increased migration and invasion.…”
