Cellular metabolism modulates effector functions in human CD4+ T (Th) cells by providing energy and building blocks. Conversely, cellular metabolic responses are modulated by various influences, e.g., age. Thus, we hypothesized that metabolic reprogramming in human Th cells during aging modulates effector functions and contributes to “inflammaging”, an aging-related, chronic, sterile, low-grade inflammatory state characterized by specific proinflammatory cytokines. Analyzing the metabolic response of human naive and memory Th cells from young and aged individuals, we observed that memory Th cells exhibit higher glycolytic and mitochondrial fluxes than naive Th cells. In contrast, the metabolism of the latter was not affected by donor age. Memory Th cells from aged donors showed a higher respiratory capacity, mitochondrial content, and intracellular ROS production than those from young donors without altering glucose uptake and cellular ATP levels, which finally resulted in higher secreted amounts of proinflammatory cytokines, e.g., IFN-γ, IP-10 from memory Th cells taken from aged donors after TCR-stimulation which were sensitive to ROS inhibition. These findings suggest that metabolic reprogramming in human memory Th cells during aging results in an increased expression of proinflammatory cytokines through enhanced ROS production, which may contribute to the pathogenesis of inflammaging.

Menopause is an important event in a woman’s life associated with hormonal changes that play a substantial role in the functioning of her body. A decline in the level of estrogens contributes to depressive symptoms and mood disorders during this period. The severity of depressive symptoms experienced by middle-aged women depends on many factors, including sociodemographic data (e.g., menopause, employment status, and marital status) and genetic variables (MAO-A and 5-HTT gene polymorphisms). In order to assess their influence on the development of depression in females, we analyzed 1453 healthy Polish women in different stages of menopause. Based on the results, we found that the l/l + l/s inheritance model for the 5-HTT gene polymorphism was more common in women without and with moderate depressive symptoms according to the Beck Depression Inventory (BDI), while the l/s model was more often observed in women with mild depression. Moreover, the overdominant 3/3 + 4/4 genotype of the MAO-A gene polymorphism was more often found in respondents without depressive symptoms, while women with depressive symptoms had more often the overdominant 3/4 genotype.

A menopausa corresponde ao último ciclo menstrual, sendo somente reconhecida depois de 12 meses, e ocorre, em média, aos 51 anos. Dessa forma, ela pode ser caracterizada como de início precoce (antes dos 40 anos) ou tardio (depois dos 55 anos). Neste cenário, as flutuações hormonais levam a alterações, não só na função reprodutiva, mas em outras importantes áreas corporais. Vale salientar que a amenorreia pode ser, também, de causa induzida, decorrente da remoção de partes do sistema reprodutivo feminino, por exemplo. Assim, a parada da atividade ovariana induz um caráter inflamatório crônico, aumentando a suscetibilidade da mulher a diversas doenças. Nessa perspectiva, o presente estudo tem como objetivo analisar as principais características da menopausa, bem como os aspectos psicológicos, o seu impacto na vida da mulher e a qualidade da assistência proporcionada pela Atenção Primária à saúde feminina. Realizou-se uma revisão integrativa da literatura nas bases de dados Medline (via PubMed) e Lilacs (via BVS), a partir da utilização de descritores (MeSH e DeCS). Aplicou-se os filtros de artigos publicados durante o período de 2015 a 2020, texto completo gratuito, pesquisas realizadas em humanos e sexo feminino. Dessa forma, foram encontrados um total de 3.548 artigos, dos quais, após a leitura dos títulos, resumos e texto na íntegra, 27 foram selecionados para compor a revisão. A menopausa pode ser assintomática ou sintomática e seus impactos levam a alterações na função reprodutiva e em outras áreas do corpo e da mente, atingindo os aspectos hormonais, imunológicos, emocionais, estressantes e sociais. O que reflete a importância da promoção à saúde ao público feminino de forma direcionada, integral, singular e de qualidade.
