Mechanism of neutrophil activation by NAF, a novel monocyte‐derived peptide agonist

Abstract: The rise in cytosolic free Ca2+, shape change, superoxide formation, and granule exocytosis induced in human neutrophils by N-formyl-Met-Leu-Phe (fMLP) and by a newly discovered activating peptide, neutrophil-activating factor, termed NAF, were compared. NAF was effective in the concentration range of 0.1-10 nM and was 10- to 100-fold more potent than fMLP. In qualitative terms, the single responses to either stimulus were remarkably similar: they showed virtually identical onset and initial kinetics, and were… Show more

“…The effects, however, are much less pronounced than those observed for neutrophils [2]. We were unable to detect [Ca*']i changes [29] or significant migration responses by human blood lymphocytes challenged with IL-8 (T. Geiser, unpublished results). Our present data show that leukemic T cells are able to migrate, but only respond to CXC chemokines when transfected with cDNAs for one of the IL-8 receptors.…”

“…It activates PMN to degranulate and exhibit respiratory burst [12,13] and is chemotactic for PMN and lymphocytes in vitro and in vivo [2,4,10,14]. Further activities include triggering of histamine and leukotriene release from IL-3-primed basophils [ 151, spasmogenic activity on airway smooth muscle [16], and effects on PMN adherence to endothelial cells [11,17].…”

“…IL-8 induces the mobilization of calcium in neutrophils (24,25), and this may occur through the activation of phospholipase C-␤ (26). There is also evidence that IL-8 stimulates phospholipase D activity (27), phosphatidylinositol-3 kinase (PI3K) activity (28), and GTP loading of RhoA (29) in human neutrophils.…”
