“…Similarly, Natalie A Sims also held the same views and believed that the JAK1/STAT3/SOCS3 axis featured in bone development, physiology and pathology (Sims, 2020). Meanwhile, recent studies have shown that mechanical stimulation improves rotator cuff tendon-bone healing by activating IL-4/JAK/STAT signaling pathway mediated macrophage-M2 polarization (Liu et al, 2022). All in all, these studies indicate that JAK/STAT pathway plays an important role in bone development and repair.…”

“…Mice were allowed for free cage activities after surgery (FC group); Mice received treadmill running initiated on postoperative day 7 (TR group); In comparision to FC group, TR group showed better fibrocartilage regeneration which was characterized by more cartilage-like cells and richer proteoglycans accumulation at repaired site at postoperative week 4 and 8. Reproduced with permission (Liu et al, 2022).…”

“…This shows that the timing of the start of exercise is very important and needs to be further defined. Running exercise has also been shown to significantly increase IL-4 production, activation of the macrophage JAK/STAT signaling pathway, macrophage polarization, and the quality of tendon bone healing (Liu et al, 2022) Figure 2F. Although all of these treatment options are effective to a certain extent, there are some limitations.…”

“…Accurate simulation of biological microstructure is important for specifically inducing injured tissue reconstruction. In fact, the TBI is a transition structure from the soft tissue to hard tissue, which can be stratified into the bone tissue, mineralized fibrocartilage, nonmineralized fibrocartilage, and tendon tissue. From the tendon to the bone, the collagen fiber gradually decreases, whereas mineral content gradually increases, thus forming a unique transition zone, which helps to uniformly transfer stress from the soft tissue to hard tissue and avoid stress concentration. , Therefore, we designed a gradient polar scaffold mimicking the transition structure of the TBI to enhance the mechanical properties of the tissue after the rotator cuff injury, while improving the inherent healing potential, thus improving the success rate of the repair of rotator cuff injury, which might be a potential tendon-bone repair strategy.…”
