Measurement of estrogenic activity in sediments from Haihe and Dagu River, China

Abstract: Sediments from two rivers in China, the Haihe and Dagu Rivers, were examined for estrogenic activity using an estrogen receptor (ER)-mediated in vitro bioassay system. ER-active compounds were isolated from sediments by Soxhlet extraction, and the crude extracts were fractionated using a florisil column into three fractions. The estrogenic activity of each extract was detected by measuring luciferase activity in the human breast cancer cell line MCF-7 transfected with a luciferase receptor gene. Significant es… Show more

“…This cell line was stably transfected with the luciferase reporter gene and estrogen-responsive element, and ER agonists can induce the production of luciferase. Estrogenic activity of each sample was determined as previously described (Song et al, 2006). Briefly, 60 interior wells of a 96-well culture plate were each seeded with 250 lL cell suspension at a density of about 5 Â 10 4 cells per well.…”

“…Industrial, domestic and agricultural wastewaters from Tianjin area are directly or indirectly discharged into the Yongding New River, Beitang River and Dagu River which carry the water to the Bohai Sea (Song et al, 2006). Samples were conducted along the rivers according to the location of chemical factories (D3), drainage outlets (B1, B2, D2), bridges (Y2), river confluence reaches (D1, Y1, Y3), and entrances of rivers (D4, Y4) (Fig.…”

“…6) BPA also has estrogenic activity and binds to estrogen receptors. 7,8) BPA might expose female health to the risk of human breast cancer. 9) First, to examine the effects of BPA on proliferation of MCF-7 cells, the cells were cultured in BPA-supplemented medium.…”
