MCMV triggers ROS/NLRP3-associated inflammasome activation in the inner ear of mice and cultured spiral ganglion neurons, contributing to sensorineural hearing loss

Abstract: Congenital cytomegalovirus (CMV) infection is the most common infectious cause of sensorineural hearing loss in children. While the importance of CMV-induced SNHL has been described, the mechanisms underlying its pathogenesis and the role of inflammatory responses remain elusive. The present study established an experimental model of hearing loss after systemic infection with murine CMV (MCMV) in newborn mice. Auditory brainstem responses were tested to evaluate hearing at 3 weeks, expression of inflammasome-a… Show more

“…ROS homeostasis was associated with neurodegeneration and auditory neuropathy [ 58 ]. Three-week-old mice infected with murine congenital cytomegalovirus (MCMV) in neonatal were found to be suffering from hearing loss, and MCMV-infected cultured SGNs in vitro displayed elevated ROS levels and activated NLRP3 inflammasome, which can be suppressed by ROS inhibitor NAC [ 59 ]. Additionally, ROS is related to cochlear neuropathy in presbycusis.…”

“…ROS production started within 30 min after infection, with a peak within 2 h [ 30 ]. In host cells, ROS are quickly produced as a defence mechanism during infections caused by HSV [ 36 , 37 ], and other members of the Herpesviridae family, such as the Epstein–Barr virus [ 38 ] and cytomegalovirus [ 39 ]. During the viral infection, ROS play a pivotal role in the activation of innate immune responses.…”

“…Histological examination showed infection, followed by a substantial loss, of the SGNs, accompanied by an infiltration of CD3+ cells and an upregulation of proinflammatory cytokines. Indeed, a prolonged inflammatory insult with high levels of IL-6, TNFα, ROS and the activation of the 'inflammasome' (Schachtele et al, 2011;Zhuang et al, 2018) may bring about the demise of the SGNs as a secondary insult rather than the primary viral infection itself, and this may be comparable to the macrophage infiltration found in the spiral ganglion region after ouabain intoxication (Brown et al, 2017).…”
