Matrix metalloproteinase distribution during early corneal wound healing

Abstract: Aim To compare matrix metalloproteinase (MMP) localisation in anterior keratectomy (AK) and lamellar keratectomy (LK) wounds. Methods Wounds were produced in one eye of 24 rabbits. The AK wounds were made to approximately 120 mm in depth and then allowed to re-epithelialise. The LK wounds were of similar depth, but the anterior stroma and epithelium were replaced after a second deeper keratectomy had been performed. Immunohistochemistry was used to localise the MMP-1, -2, -3, and -9 at intervals from 4 h to 14… Show more

“…Fifty nine sections excised 4mm from the limbus, that were not fragmented or crushed were selected because trauma can release MMPs into the extracellular space, 13 which could be mistaken to cause pterygium remodeling. 5,7 Seven control sections were obtained from the nasal corneo-conjunctiva of patients that were undergoing evisceration for irreparably injured eyes.…”

“…Peroxynitrite and NO reduce the levels of the tissue inhibitor of metalloproteinase-1 (TIMP-1), increasing the gelatinolytic activity of MMPs in corneal fibroblasts cultivated in vitro (11) . MMP-9 is produced by epithelial cells and neutrophils and has a role in remodeling the corneal stroma after keratectomy (12) . MMP-9 is involved in corneal stroma degradation, facilitating the migration and proliferation of neo-vessels in ulcers (12) .…”
