Marrow Adipose Tissue: Its Origin, Function, and Regulation in Bone Remodeling and Regeneration

Abstract: Marrow adipose tissue (MAT) is a unique fat depot in the bone marrow and exhibits close relationship with hematopoiesis and bone homeostasis. MAT is distinct from peripheral adipose tissue in respect of its heterogeneous origin, site-specific distribution, and complex and perplexing function. Though MAT is indicated to function in hematopoiesis, skeletal remodeling, and energy metabolism, its explicit characterization still requires further research. In this review, we highlight recent advancement made in MAT … Show more

“…Bone marrow adipose tissue (BMAT) thus refers to BM areas where BMAds are the predominant cell type, and BMA refers more broadly to BMAT across all skeletal locations and metabolic states. Over the last decades, interest in the functional role of BMAds has gradually increased and it is now evident that BMAds are actively involved in bone metabolism, hematopoiesis, and energy metabolism (1,2). In addition, a possible role for BMAds in many diseases has emerged (3), and research groups all over the world are investigating the origin, function, and interaction of BMAds.…”

“…Conversely, PTH1R deletion in mesenchymal stem cells (MSCs) led to formation of adipocytes ( Fan et al, 2017 ), and mice treated with Bisphenol-A-diglycidyl ether, a PPARγ antagonist, showed higher levels of osteoblastogenesis and bone formation concomitant with decreased marrow adiposity and ex vivo adipogenesis ( Duque et al, 2013 ). However, no existing evidence can directly prove a causal relationship between BMA and bone loss ( Li, Wu & Kang, 2018 ). Therefore, further investigations of intrinsic mechanism underlying adipogenesis of BMSCs at an early stage may provide new insight into BMSCs lineage commitment.…”

“…BMAT is thought to be derived from bone marrow stromal (skeletal, mesenchymal) stem cells (BMSC) present within the bone marrow stroma and that are capable for differentiation, in addition to adipocytes, into osteoblasts and chondrocytes [1]. Currently, there is no consensus regarding the phenotype of BMAT progenitors but a number of markers have been proposed to identify adipocyte progenitors within the bone marrow, including osterix [4,5], Prx1 and Nestin1 (reviewed in [6]), leptin receptor [7], Rankl [8], Znf423 [9••], and Hoxa11 [10].…”
