Mammalian cell cytotoxicity and genotoxicity analysis of drinking water disinfection by‐products

Abstract: Cytotoxicity and genotoxicity assays were used to analyze drinking water disinfection by-products (DBPs) in Chinese hamster ovary (CHO) AS52 cells. The DBPs were chosen because they are common in drinking water, resulting from conventional disinfection using chlorination and chloramination. Data were also available to compare these results with cytotoxicity and mutagenicity studies in Salmonella typhimurium. The rank order in decreasing chronic cytotoxicity measured in a microplate-based assay was bromoacetic … Show more

“…Newly formed Br-DBPs were clearly observed in the mass spectrum (Figure 1c). Similar to the previously detected chlorinated DBPs, 12,13 Br-DBPs also have a characteristic mass difference of 36.4 mDa for the replacement of O by CH 4 . In total, molecular formulas of 441 peaks containing one bromine and 37 peaks containing two bromines were identified in this study.…”

“…The observation of decarboxylation is in agreement with previous fragmentation studies of fulvic acid. 14,22,37 However, losses of H 2 O, CO, or CH 4 , which have been previously reported in MS−MS analysis of fulvic acid, 14,22 were not observed. This difference may be attributed to the differences in the structures of the precursor ions.…”

“…After the gel hardened, a second layer of 0.5% low melting point agarose was applied. Detailed methods for preparing and electrophoresing the SCGE microgels were published previously (50)(51)(52). Cellular membranes were removed by an overnight immersion in lysing solution at 4jC.…”

“…Many of these DBPs have been shown to be cytotoxic, genetoxic or cause cancer in laboratory animals, and for 44 these reasons, they are considered to be of public health concern (Plewa et al 2002). Some epidemiological studies have …”
