Abstract: Epigenetic alterations have been reported to play critical roles in the development of colorectal cancer (CRC). However, the biological function of the lysine-specific histone demethylase 1B (LSD2/KDM1B) in CRC is not well understood. Therefore, we investigated the characteristics of LSD2 in CRC. We observed significant upregulation of LSD2 in CRC tissue compared to that in normal colorectal tissue. LSD2 promotes CRC cell proliferation and inhibits cell apoptosis through cell cycle regulation, promoting CRC pr… Show more

“…Alterations in histone methylation have a global influence on drug resistance in ovarian cancer cells ( 66 , 67 ). In the present study, reduction of histone H3k4me2 methylation was observed in CRC cells and attributed to GS-induced chemoresistance to 5FU therapy, in accordance with a previous study which showed that H3k4me2 demethylation played an important role in CRC progression ( 68 ). In gastric cancer, LSD1 specifically catalyzed the demethylation of mono- and di-methylated H3k4me2, participating in many pathological processes of cancer, including proliferation, apoptosis, and metastasis ( 69 ).…”

“…Moreover, LSD2 downregulates p53 expression through H3K4me2 demethylation in the p53 promoter region, thus driving the cell cycle through p53-p21-Rb. In fact, LSD2 is associated with a decrease in p53 and p21 protein levels and an increase of Rb protein levels [44].…”

“…Increased expression of ADPGKAS1 and LSD2 may be directly related to the PI3K/AKT/mTOR signalling pathway in GC tissues [11]. In addition, ADPGKAS1 downregulates p53 through the regulation of LSD2 [44] There is little literature regarding the role of LSD1 and LSD2 in GC (Table 1), and the main conclusions are summarised in Figure 1.…”

“…Histone lysine-specific demethylases 1 and 2 (LSD1, LSD2) are demethylases that bind to p53 and demethylate K370me2 at Lys370 (LSD1) or K3K4me2 in the TP53 promoter (LSD2). This leads to the inhibition of p53 function and p53-mediated apoptosis [84][85][86].…”
