Abstract: High-risk human papillomaviruses (HR HPV) cause cervical cancer, and in these cancers, HPV type 16 is the most common HR type. The HR viral oncogenes E6 and E7 partner with cellular proteins to drive cancer and modulate immune pathways; previously, we demonstrated in keratinocytes that HPV 16 E6 and high expression of the endogenous host protein partner NFX1-123 led to the increased expression of multiple genes, including Notch1, secretory leukocyte peptidase inhibitor (SLPI), and retinoic acid early transcrip… Show more

“…High expression of NFX1-123 has been demonstrated in HPV-positive cervical cancer cell lines [ 41 ] and in primary cervical cancers, which are nearly universally HPV-positive [ 39 , 43 ]. In normal keratinocytes expressing 16E6, greater expression of NFX1-123 was associated with extended longitudinal active cellular growth and augment hTERT expression along with telomerase activity [ 39 ].…”

“…In our recent studies, we confirmed NFX1-123 expression in HPV-positive HNSCCs was higher than that found in HPV-negative HNSCCs (manuscript in preparation). This indicated that, like in cervical cancer, NFX1-123 may have a specific significance in HPV-positive HNSCCs, and its high expression functions in a manner similar to that in cervical cancer [ 39 , 43 ].…”

“…3.1 | NFX1-123, but not NFX1-91, was increased in human papillomavirus positive head and neck squamous cell carcinomas NFX1-123 was previously identified as a protein partner of 16E6, and its expression is increased in cervical cancer cell lines and primary cervical cancers. [16][17][18] There are three splice variants of the NFX1 gene in NCBI, and two are expressed at the protein level in epithelial cells and keratinocytes. Those two splice variants, NFX1-123 and NFX1-91, have been the focus of studies in cervical cancers, but have not been studied in HNSCCs.…”

“…23 We have recently confirmed that many genes upregulated by NFX1-123 and 16E6 in primary human foreskin keratinocytes (HFKs) were also overexpressed in cervical cancers when compared to the normal cervix. 17 This indicates that NFX1-123 plays a functional role in cervical cancer, and it also suggests that NFX1-123 may be important in HPV+ cancers at other anatomic sites.…”
