Anti-MAdCAM-1-Conjugated Nanocarriers Delivering Quantum Dots Enable Specific Imaging of Inflammatory Bowel Disease

Abstract: Purpose Assessment of inflammatory bowel disease (IBD) currently relies on aspecific clinical signs of bowel inflammation. Specific imaging of the diseased bowel regions is still lacking. Here, we investigate mucosal addressin cell adhesion molecule 1 (MAdCAM-1) as a reliable and specific endothelial target for engineered nanoparticles delivering imaging agents to obtain an exact mapping of diseased bowel foci. Materials and Methods We generated a nanodevice composed of… Show more

“…These NPs were intravenously injected into IBD mice. The T1-weighted images obtained after 24 h showed enhancement in the rectal mucosa, proving the specific localization of PLGA NPs [ 107 ]. This technique mimics the behavior of circulating leukocytes during inflammation and allows for the tracking of their interaction with adhesion molecules whose expression is enhanced during inflammatory conditions.…”

“…Imaging has long been utilized to evaluate segments of the digestive tract that are inaccessible with conventional endoscopy. For decades, nanoparticles such as Au, 116 , 117 Fe and Au complexes, 118 quantum dots, 119 , 120 MnO, 82 and lipid nanoparticles 74 have been developed and used in cancer imaging and diagnosis. During traditional clinical CT imaging, iodinated small molecules and barium sulfate suspensions are usually used as contrast agents, 121 , 122 but they are not ideal for imaging IBD due to their nonspecificity.…”

“…In another study, Fabio Corsi and coworkers loaded quantum dots into anti-MAdCAM-1 coupled PLGA-PEG nanoparticles (P@QD-MdC NPs). 120 Upon intravenous injection, the system is expected to specifically bind to MAdCAM-1 protein on the apical side of mucosal endothelial vessels and penetrate the bowel mucosa. In vivo experiments showed progressive P@QD-MdC NP accumulation in the inflamed colon and provided an enhanced and localized signal.…”

“…In the context of nanoparticles properly designed for the use as MRI contrast agents, it is worth mentioning the work of Truffi et al [ 43 ]. They prepared poly(lactic-co-glycolic acid)-block-b-poly(ethyleneglycol) (PLGA-PEG) polymer micelles targeted to the endothelial mucosal addressin cell adhesion molecule 1 (MAdCAM-1), conjugated with the half-chain of a monoclonal MAdCAM-1 antibody.…”
