Abstract: Lysyl oxidase-like 2 (LOXL2), a member of the lysyl oxidase gene family, is involved in the progression of hepatocellular carcinoma progression and metastasis. Increased expression of LOXL2 has been identified in several types of cancer, including hepatocellular carcinoma. Recently, LOXL2 has been reported to promote epithelial-mesenchymal transition by reducing E-cadherin expression via the upregulation of Snail expression. The present study provided evidence demonstrating that LOXL2 inhibited the expression … Show more

“…In addition to LOXL2′s effect on non-cellular components, recent studies have demonstrated its role in boosting angiogenesis through intercellular crosstalk. As such, Fan et al demonstrated LOXL2 activates the SNAIL/fructose-1,6-biphosphatase (FBP1) axis to upregulate the HIF-1α/VEGF pathway in HCC cells, leading to enhanced tubular networks of endothelial cells [ 49 ]. Xing et al reported that increased matrix stiffness can induce M2 polarization of the THP-1 cell line and increase LOXL2 expression through activation of integrin β5- focal adhesion kinase (FAK), mitogen-activated protein kinase kinase (MEK), 1/2-extracellular signal-regulated kinase (ERK) 1/2 pathway and upregulation of HIF-1α [ 50 ].…”

“…Moreover, Wong et al showed that the TME-fostering effect of LOXL2 relies on the activation of the HIF-1α/TGF-B/SMAD4 pathway [ 42 ]. Interestingly, LOXL2 and HIF-1α are thought to exert reciprocal modulation; as such, Fan et al showed that LOXL2 can act to upregulate HIF-1α through the Snail-FBP1 axis [ 49 ]. In addition, the forkhead box M1b (FoxM1b), a transcription factor which exerts a pro-tumor effect on HCC, has been reported to play a positive role in LOX and LOXL2 expression.…”

“…A total of 1452 proteins were quantified of which 107 were found to be differentially expressed (DE) in PQ-treated EVs (│FC│ > 1.2; LFDR ≤ 0.05) ( Figure S2 and Table S3 ). As illustrated in the volcano plot ( Figure S2 ), among the most highly DE proteins of the 107 DE proteins, three (LOXL2, MMP2 and PHB2) are known to have a link with hypoxia response [ 37 , 38 , 39 ]. This observation permits to denote a previously described effect of the PQ induction of hypoxia-related pathways [ 40 ].…”

“…LOXL2 is regulated by hypoxia and can promote the hypoxia pathway. In liver cancer, HIF-1α promotes LOXL2 expression, which promotes EET and VM through the Snail/FBP1/VEGF pathway [143]. The tumor suppressor protein p53induced nuclear protein 1 (TP53INP1) plays an inhibitory role in tumor invasion and metastasis and has been shown to inhibit hypoxia-induced EET through the ROS/GSK-3beta/Snail pathway [144].…”
