Abstract: OBJECTIVEThe NLRP3 inflammasome plays an important regulatory role in obesity-induced insulin resistance. NSAID activated gene-1 (NAG-1) is a divergent member of the TGF-β superfamily. NAG-1 Tg mice are resistant to dietary- and genetic-induced obesity and have improved insulin sensitivity. Our objective was to examine whether NLRP3 inflammasome activity is associated with this observed phenotype in NAG-1 Tg mice.DESIGN AND METHODSKey components of the NLRP3 inflammasome were examined in NAG-1 Tg mice on both … Show more

“…The hNAG-1 mice have most of the metabolic characteristic of long-lived mice [1]. In addition, we have previously reported that the hNAG-1 mice have a lower inflammatory response [15, 16], and inflammation has been associated with aging [28, 29]. In contrast to most long-lived animals in which genetic deletion increases life span, we shown here that an increase in the expression of hNAG-1 protein extended longevity.…”

“…Recent evidence suggests that aberrant activation of NLRP3 inflammasome, an intracellular inflammatory machinery, is involved in the development of different chronic degenerative diseases and pathologic processes. Examples include diabetes mellitus (Masters et al, 2010;Grishman et al, 2012;Lee et al, 2013), obesity (Esser et al, 2013;Wang et al, 2014), silicosis (Peeters et al, 2013;Peeters et al, 2014), liver cirrhosis (Wree et al, 2014), and ESRD (Abais et al, 2014a, b;Xia et al, 2014). The latter group of studies by our laboratory demonstrate that the formation and activation of the NLRP3 inflammasome contributes to glomerular injury from a sterile inflammatory response to hHcys.…”
