Abstract: Glycosylation of immunoglobulin G (IgG) influences IgG effector function by modulating binding to Fc receptors. To identify genetic loci associated with IgG glycosylation, we quantitated N-linked IgG glycans using two approaches. After isolating IgG from human plasma, we performed 77 quantitative measurements of N-glycosylation using ultra-performance liquid chromatography (UPLC) in 2,247 individuals from four European discovery populations. In parallel, we measured IgG N-glycans using MALDI-TOF mass spectrome… Show more

“…20,21 Some hepatocyte-produced glycoproteins and their glycosylated forms have been intensively studied and used as disease markers. 12,13,22 Among these proteins, a core-fucosylated heterogeneous form of AFP (AFP-L3) is used as a HCC surveillance marker. 13 Although false positivity of serum AFP in HCC surveillance was partially improved by combined diagnosis of AFP and AFP-L3, there remains a 30-40% chance of false-negative findings using this combined marker in HCC identification.…”

“…In an important class of GWAS (analysis of "omics" phenotypes), the studies involve many traits y j [12,13,14,15]. In this case, the set of generalized least squares problems in Eq.…”

“…Protein glycosylation is involved in many key cellular and physiological processes, [14] and can furthermore provide a cell with a mechanism for rapid adaptation by integration of environmental information. [15] Large-scale genome-wide association studies (GWAS) [16,17] have recently identified many putative loci associated with protein glycosylation. Two of them, the HNF1A gene (encoding the hepatic nuclear factor alpha) and the IL6ST gene (the interleukin 6 signal transducer shared by many cytokines) are particularly interesting in our studies of epigenetic regulation of protein glycosylation in complex diseases.…”

“…HNF1A-MODY). [18] On the other hand, the IL6ST has been associated both with immunoglobulin G (IgG) glycosylation and inflammatory and autoimmune conditions, such as inflammatory bowel disease (IBD), [17] which we are currently studying in terms of epigenetics and glycosylation. [19] The aim of this work was to assemble several genome editing tools for knockout of human genes HNF1A and IL6ST and select the most efficient TALEN or CRISPRCas9 construct for each gene using the luciferase assay.…”
