Acetylcholine (ACh) signaling orchestrates mammalian movement, mental capacities, and inflammation. Dysregulated ACh signaling associates with many human mental disorders and neurodegeneration in an individual-, sex-, and tissue-related manner. Moreover, aged patients under anticholinergic therapy show increased risk of dementia, but the underlying molecular mechanisms are incompletely understood. Here, we report that certain cholinergictargeting noncoding RNAs, named Cholino-noncoding RNAs (ncRNAs), can modulate ACh signaling, agonistically or antagonistically, via distinct direct and indirect mechanisms and at different timescales. Cholino-ncRNAs include both small microRNAs (miRNAs) and long noncoding RNAs (lncRNAs). The former may attenuate translation and/or induce destruction of target mRNAs that code for either ACh-signaling proteins or transcription factors controlling the expression of cholinergic genes. lncRNAs may block miRNAs via 'sponging' events or by competitive binding to the cholinergic target mRNAs. Also, single nucleotide polymorphisms in either Cholino-ncRNAs or in their recognition sites in the ACh-signaling associated genes may modify ACh signaling-regulated processes. Taken together, both inherited and acquired changes in the function of Cholino-ncRNAs impact ACh-related deficiencies, opening new venues for individual, sex-related, and age-specific oriented research, diagnosis, and therapeutics.

in recent years, an increasing number of long non-coding rnas (lncrnas) have been discovered using microarrays and nucleic acid sequencing technology. lncrnas exert crucial biological functions by regulating signaling pathways. In particular, the lncRNA growth arrest-specific transcript 5 (GaS5) has been documented to serve a crucial role in numerous signaling pathways. This article discusses the latest developments in the association between GaS5 and microrna (mirna), p53, mTor, glucocorticoid response element (Gre) and aKT in order to investigate the roles served by GaS5. mirnas can activate related signaling pathways and GaS5 can combine with mirna to regulate related signaling pathways. GaS5 may regulate p53 expression via derivation of snorna, but the underlying mechanism requires further investigation. GaS5 overxpresion reduces the expression level of mTor, which is induced by inhibiting mir-106a-5p expression. GaS5 is a sponge of Gr, and serves a role in controlling and maintaining glucocorticoid sensitivity and drug resistance via competitive combination with Gr. GaS5 can interact with mirnas, such as mir-21 and mir-532-5p, to regulate the expression of aKT signaling pathway, affecting cell survival and apoptosis. collectively, the data indicate that GaS5 serves a key role in the mirna, p53, mTor, Gre, and aKT signaling pathways. GaS5 regulates complex intracellular signaling pathways primarily through three modes of action, all of which are interrelated: Signal, decoy and guide. in the present article, latest developments in the association between GaS5 and a number of cellular signaling pathways are discussed to examine the tumor suppressive role of GaS5. Renal fibrosis is an unavoidable consequence that occurs in nearly all of the nephropathies. It is characterized by a superabundant deposition and accumulation of extracellular matrix (ECM). All compartments in the kidney can be affected, including interstitium, glomeruli, vasculature, and other connective tissue, during the pathogenesis of renal fibrosis. The development of this process eventually causes destruction of renal parenchyma and end-stage renal failure, which is a devastating disease that requires renal replacement therapies. Recently, long non-coding RNAs (lncRNAs) have been emerging as key regulators governing gene expression and affecting various biological processes. These versatile roles include transcriptional regulation, organization of nuclear domains, and the regulation of RNA molecules or proteins. Current evidence proposes the involvement of lncRNAs in the pathologic process of kidney fibrosis. In this review, the biological relevance of lncRNAs in renal fibrosis will be clarified as important novel regulators and potential therapeutic targets. The biology, and subsequently the current understanding, of lncRNAs in renal fibrosis are demonstrated—highlighting the involvement of lncRNAs in kidney cell function, phenotype transition, and vascular damage and rarefaction. Finally, we discuss challenges and future prospects of lncRNAs in diagnostic markers and potential therapeutic targets, hoping to further inspire the management of renal fibrosis.
