Abstract: Abstract.A high performance liquid chromatographic method has been developed for the resolution ofpenicillamine enantiomers on a/~-cyclodextrin (/%CD) column. Fluorogenic derivatives of the D,L-penicillamine were first formed by using o-phthaldehyde/2-mercaptoethanol (OPA/2-ME) as derivatizating reagent. These chiral derivatives were then chromatographed on a commercially available chiral stationary phase of/%CD with 50/50 ethanol/l% triethylammonium acetate (pH 4.5) as a mobile phase, and detected fluorimetri… Show more

“…A fl uorescent chiral reagent, 4-(3-isothiocyanato-pyrrolidin-1-yl)-7-(N,N-dimethyl-aminosulfonyl)-2,1,3 benzoxadiazole (DBD-PyNCS), has been used for indirect HPLC resolution on an ODS column (Jin and Toyo'oka, 1998) while the fl uorescent tagging reagent, N-(1-pyrenyl) maleimide (NPM), has been used for direct resolution on Teicoplanin and b-CD columns (Kullman et al, 2000). Enantiomers of PenA derivatized with OPA and 2-mercaptoethanol were separated on a b-CD column (Merino et al, 1992), but sensitivity was low in spite of fl uorometric detection.…”

“…These phases were used with both non-polar eluents and aqueous systems in the presence of Cu 2+ ions in the ligandexchange mode. Isoindole adduct derivatives prepared with OPA, 2-mercaptoethanol (Merino et al, 1992) were separated using HPLC on b-CD column. The method was also applied to determine the enantiomeric purity of D-PenA in pharmaceutical tablet forms.…”

“…OPhthalaldehyde (Duchateau et al, 2004) and a fluorescent chiral tagging reagent 4-(3-isothiocyanatopyrrolidin-1-yl)-7-(N,N-dimethylaminosulfonyl)-2,1,3-benzoxa-diazole (DBD-PyNCS) has been used for indirect resolution with an ODS column (Jin and Toyo'oka, 1998), while the fluorescent tagging reagent N-(1-pyrenyl)maleimide (NPM) has been used for direct resolution with teicoplanin and β-CD columns (Kullman et al, 2000). PenA enantiomers derivatized with OPA and 2-mercaptoethanol were separated on the β-CD column (Merino et al, 1992). TLC provides direct resolution of enantiomers of a variety of compounds and is used for its several advantages (Bhushan and Martens, 2003).…”

“…For example, enantiomers of penicillamine have different pharmacological effects, and only u-penicillamine can be used in the treatment of Wilson's disease, polyarthritis cystinuria and heavy metal poisoning (Crawhall et af., 1979). Recently, penicillamine enantiomers derivatized with ophthalaldehyde and 2-mercaptoethanol (OPA/2-ME) were separated on a P-cyclodextrin column (Merino et al, 1992). The method is simple and rapid, but the sensitivity was low in spite of fluorornetric detection.…”
