Dopaminergic modulation of dye coupling between neurons in the core and shell regions of the nucleus accumbens

The presence of dye coupling and electrotonic coupling within the nucleus accumbens was examined using intracellular recording and staining in rat brain slices. In 24% of the cells examined, injection of single accumbens neurons with the dye Lucifer yellow resulted in the complete labeling of more than one cell. To examine whether this coupling is regulated by dopamine (DA), the extent of dye coupling was assessed in the presence of dopaminergic agonists and antagonists. In the core region of the accumbens, D1 receptor stimulation induced a decrease in the incidence of dye coupling; this effect could be blocked by the D1 antagonists SCH 23,390 and SCH 39,166, by the D2 antagonist sulpiride, or by depletion of DA by reserpine and alpha-methyl-p- tyrosine coadministration, but not by the atypical antipsychotic drug clozapine. In contrast, in the posterior part of the shell region, D2 receptor stimulation increased the incidence of coupling independent of the state of D1 receptor activation. Fast spikelets resembling coupling potentials were found to occur only in neurons that also showed dye coupling, and could be attenuated in core neurons by D1 receptor stimulation. These findings suggest that DA, in addition to its actions on the membrane of single neurons, may also affect neuronal integration on a network level by regulating the transfer of information between adjacent neurons within the nucleus accumbens.

Because degranulation of brain mast cells activates adrenocortical secretion ( 41 , 42 ), we examined whether activation of such cells increases renin and vasopressin (antidiuretic hormone: ADH) secretion. For this, we administered compound 48/80 (C48/80), which liberates histamine from mast cells, to pentobarbital-anesthetized dogs. An infusion of 37.5 μg/kg C48/80 into the cerebral third ventricle evoked increases in plasma renin activity (PRA), and in plasma epinephrine (Epi) and ADH concentrations. Ketotifen (mast cell-stabilizing drug; given orally for 1 wk before the experiment) significantly reduced the C48/80-induced increases in PRA, Epi, and ADH. Resection of the bilateral splanchnic nerves (SPX) below the diaphragm completely prevented the C48/80-induced increases in PRA and Epi, but potentiated the C48/80-induced increase in ADH and elevated the plasma Epi level before and after C48/80 challenge. No significant changes in mean arterial blood pressure, heart rate, concentrations of plasma electrolytes (Na+, K+, and Cl−), or plasma osmolality were observed after C48/80 challenge in dogs with or without SPX. Pyrilamine maleate (H1histaminergic-receptor antagonist) significantly reduced the C48/80-induced increase in PRA when given intracerebroventricularly, but not when given intravenously. In contrast, metiamide (H2histaminergic-receptor antagonist) given intracerebroventricularly significantly potentiated the C48/80-induced PRA increase. A small dose of histamine (5 μg/kg) administered intracerebroventricularly increased PRA twofold and ADH fourfold (vs. their basal level). These results suggest that in dogs, endogenous histamine liberated from brain mast cells may increase renin and Epi secretion (via the sympathetic outflow) and ADH secretion (via the central nervous system).

Male and female Sprague-Dawley rats were treated with nicotine during the prenatal period and the first three postnatal weeks (the pregnant and lactating rats were given nicotine hydrogen ( + ) tartrate (165 mg/l) in the tap water). Catecholamine fluorescence was evaluated using quantitative histofluorometry on brain sections treated according to Falck-Hillarp methodology. In order to evaluate catecholamine utilization in discrete hypothalamic catecholamine nerve terminal networks, the aMT-(a-methyl-( f)-p-tyrosine methyl ester) induced catecholamine disappearance was studied 2 h following the tyrosine hydroxylase inhibition. The body weight was reduced in both the male and female rats from 3 weeks of age until 9 weeks of age following pre-and postnatal treatment with nicotine.Following one week of withdrawal from pre-and postnatal treatment with nicotine, an increased catecholamine utilization was observed in the medial and lateral palisade zones of the median eminence mainly in the female rat. In the female rat, reduced prolactin serum levels were found both in the presence and absence of aMT treatment as well as reduced luteinizing hormone concentration in the presence of aMT treatment.At 6 months of age indications of a maintained, weak activation of the catecholamine nerve terminal systems in the medial palisade zones of the median eminence were observed in male rats pre-and postnatally treated with nicotine. Furthermore, increased noradrenaline levels were found in the paraventricular hypothalamic nucleus. An increase in serum luteinizing hormone levels was also found in these rats. In the 7-month old diestrous rat, maintained marked increases in catecholamine utilization in the medial and lateral palisade zones of the median eminence were found following treatment with nicotine during the pre-and postnatal period. A significant reduction of nigral dopamine stores was also demonstrated. The serum levels of thyroid stimulating hormone, prolactin and luteinizing hormone were unchanged in these rats both in the presence and absence of tyrosine hydroxylase inhibition. Finally, pre-and postnatal treatment with nicotine did not alter 13H]nicotine binding (quantitative receptor autoradiography) in cortical, striatal and thalamic areas of the adult diestrous rat.The results demonstrate that pre-and postnatal treatment with nicotine in the drinking water produces permanent activations of the catecholamine nerve terminal networks of the external layer of the median eminence mainly in the female rat. These changes appear to be associated with reduced serum prolactin levels in the 4-week old female rat. Sex-specific changes occur in discrete noradrenaline nerve terminal systems. The observed changes may have functional consequences for neuroendocrine regulation and for the regulation of food and water intake as well as sex-specific responses to stress in the male versus the female rat.
